DescriptionAGAP2 (also known as PIKE-A), a putative oncogene, shows elevated expression in cancer cells compared to non-transformed cells and has been implicated as a regulator of cancer cell invasion into normal tissues. A contributor to brain, eye, breast and liver cancers, AGAP2 belongs to the centaurin gamma-like family with two ANK repeats, an Arf-GAP domain, a Miro domain and a PH domain. It is a GTPase-activating protein (GAP) for ARF1 and ARF5, which also shows strong GTPase activity. The Arf-GAP domain interacts with the G domain and may regulate its GTPase activity. GAP activity of AGAP2 is stimulated by phosphatidylinositol 4,5-bisphosphate (PIP2) and, to a lesser extent, by phosphatidylinositol 3,4,5-trisphosphate (PIP3). Phosphatidic acid potentiates PIP2 stimulation. Several isoforms of AGAP2 have been reported.
