The melanocortins are regulatory neuropeptidesderived by post-translational processing of the larger 30 kDa, pro-opiomelanocortin (POMC) precursor, and include the adrenocorticotropic hormone (ACTH) and the melanocyte stimulating hormones (α-, β-, and γ-MSH). The melanocortins are produced primarily in the anterior and intermediate lobes of the pituitary, in the arcuate nucleus of the hypothalamus, and to a lower level in various other peripheral tissues. In addition to their well established melanotropic and adrenocorticotropic actions, these peptides have been reported to exert a broad variety of physiological actions including those related to central neural function such as behavior, memory, cognition, and regulation of feeding. Peripherally, melanocortins have immunomodulatory and neurotrophic properties.A suggested positive control for IHC is Bouin-fixed paraffin or Stefanini-fixed frozen sections of pig pituitary. Over-night incubation at 4ºC is recommended.Reconstitute with 50-100 ul of distilled water, and dilute further in 0.1M PBS with 1% BSA and 0.1% sodium azide. For long-term storage, aliquot and store at -20ºC.
