A suggested positive control for IHC is Bouin-fixed paraffin or Stefanini-fixed frozen sections of pig pituitary. Over-night incubation at 4°C is recommended. Reconstitute with 50-100 µL of distilled water, and dilute further in 0.1M PBS with 1% BSA and 0.1% sodium azide. For long-term storage, aliquot and store at -20°C.The melanocortins are regulatory neuropeptidesderived by post-translational processing of the larger 30 kDa, pro-opiomelanocortin (POMC) precursor, and include the adrenocorticotropic hormone (ACTH) and the melanocyte stimulating hormones (?-, ?-, and ?-MSH). The melanocortins are produced primarily in the anterior and intermediate lobes of the pituitary, in the arcuate nucleus of the hypothalamus, and to a lower level in various other peripheral tissues. In addition to their well established melanotropic and adrenocorticotropic actions, these peptides have been reported to exert a broad variety of physiological actions including those related to central neural function such as behavior, memory, cognition, and regulation of feeding. Peripherally, melanocortins have immunomodulatory and neurotrophic properties.
