PA1-317 detects CBF-beta in human samples. PA1-317 has been successfully used in Western blot procedures. By Western blot, this antibody detects a ∽22 kDa protein representing CBF-beta in HeLa whole cell lysates. The PA1-317 immunogen is a synthetic peptide corresponding to residues M(1) P R V V P D Q R S K F E N E E F F R K(20) of human CBF beta. This sequence is conserved in mouse. PA1-317 immunizing peptide (Cat. No. PEP-282) is available for use in neutralization and control experiments.

CBF-beta (Core-binding factor beta), also called polyomavirus enhancer binding protein 2 beta (PEBP2B), belongs to the PEBP2/CBF transcription factor family which regulates a host of genes specific to hematopoiesis (e.g., RUNX1) and osteogenesis (e.g., RUNX2). CBF-beta is associated with an inversion of chromosome 16 and is associated with acute myeloid leukemia in humans. CBFbeta forms a heterodimer with RUNX1 (runt-related transcription factor 1), which has a DNA binding domain homologous to the pair-rule protein runt in Drosophila melanogaster. Both RUNX1 and CBFbeta are essential for hematopoiesis. Haploinsufficiency of another runt-related protein, RUNX2 (also called CBFA1), causes cleidocranial dysplasia in humans and is essential in skeletal development by regulating osteoblast differentiation and chondrocyte maturation. CBF-beta is expressed in all tissues tested. CBF-bet is highly expressed in the thymus, but also abundantly expressed in muscle, lung and brain.
