MA1-40064 detects CD14 from mouse samples. MA1-40064 has been successfully used in flow cytometry, functional assays, and immunofluorescence applications. The MA1-40064 immunogen is mouse monocyte marker CD14. Endotoxin <24 EU/mg. Positive control: RAW cells.

CD14 is a 55 kDa GPI-anchored glycoprotein that is constitutively expressed on the surface of mature monocytes, macrophages, and neutrophils. The CD14 gene consists of two exons which code for a single mRNA that is translated into a protein of 375 amino acids. The CD14 protein is composed of eleven leucin-rich repeats, which are also found in TLR and which are important in PAMP binding. In contrast to TLR, however, CD14 lacks a transmembrane domain, and thus cannot initiate intracellular signal transduction by itself. The most important CD14 signaling co-receptor is toll-like receptor 4 (TLR4), which activates, among others, the nuclear factor ?B (NF-?B) inflammatory pathway. CD14 also serves as a multifunctional lipopolysaccharide receptor, and is released to the serum both as a secreted and enzymatically cleaved GPI-anchored form. CD14 binds lipopolysaccharide molecule in a reaction catalyzed by lipopolysaccharide-binding protein (LBP), an acute phase serum protein. The soluble sCD14 can discriminate slight structural differences between lipopolysaccharides and is important for neutralization of serum allochthonous lipopolysaccharides by reconstituted lipoprotein particles. Further, CD14 has been shown to bind apoptotic cells, and can affect allergic, inflammatory and infectious processes. Alternative splicing results in multiple transcript variants encoding the same CD14 isoform. Diseases associated with CD14 dysfunction include mycobacterium chelonae infection and Croup.
