PA1-028 detects cyclophilin D (CyPD) in rat, hamster, canine, mouse, and human tissues. PA1-028 has been sucessfully used in Western blot procedures. By Western blot, this antibody detects a ~15 kDa protein in rat liver and heart extract, and a ~37 kDa in rat brain extract. The PA1-028 immunogen is a synthetic peptide corresponding to residues S(42) G N P L V Y L D V D A N(54) C of mature human CyPD. This peptide (Cat. No. PEP-104) is available for use in neutralization and control experiments.Cyclophilin D (CyPD) is a member of the immunophilin family of proteins. Immunophilins are a family of soluble cytosolic receptors capable of binding to one of two major immunosuppressant agents - cyclosporin A (CsA) or FK506. Proteins which bind FK506 are termed FK506 Binding Proteins (FKBP ft.s) and those which bind cyclosporin A are called cyclophilins (CyP). Immunophilins function as peptidyl prolyl cis-trans-isomerases (PPIase) whose activity is inhibited by their respective immunosuppressant compounds. As PPIase ft.s, immunophilins accelerate folding of some proteins both in vivo and in vitro by catalyzing slow steps in the initial folding and rearrangement of proline containing proteins. CyPD is located in the inter-membrane space of the mitochondria where it has been shown to be an integral member of the permeability transition pore complex which also includes the voltage dependent anion channel (VDAC) and the adenine nucleotide translocator (ANT).
