Description
Chronic exposure to interferons inhibits cell proliferation and results in cell death. The gene for the Death Associated Protein-3 (DAP3) was identified using a novel approach named Technical Knockout. Briefly, HeLa cells were tranfected with an antisense cDNA expression library, then exposed to interferon-γ. Surviving cells were rescued and the anti-sense “protecting” genes were isolated. Several genes were identified, including one for DAP3. The DAP3 gene was found widely expressed and, as predicted, DAP3 antisense mRNA protected HeLa cells from the killing effects of interferon-γ. Thus, this novel protein lies somewhere in the signaling downstream of interferon-γ induced cell death.
