Fas/APO-1 was originally described as a cell surface antigen on activated human T and B lymphocytes and a variety of malignant human lymphoid cell lines. The gene encoding the human fas antigen consists of 9 exons, spans more than 26 kb of DNA and shares 58.5% homology with the mouse homolog. The protein contains a single transmembrane domain which is rich in cysteine residues and an inhibitory as well as a signal transducing or death domain in the cytoplasmic domain. Ligation or crosslinking of the extracellular domain by fas ligand or by antibody elicits a characteristic apoptotic response in susceptible cells including lymphoid cells and tumors as well as non-hematopoietic tumors. Upon activation, fas associates with a number of other proteins to mediate the response. Fas is a member of the TNF receptor family and shares some sequence homology with the TNF receptor but does not appear to physically interact with TNF-R itself.
