Extracellular ATP acts as a signaling molecule to mediate a wide variety of physiological responses through interactions with the P2 family of receptors. P2 receptors have been classified into two groups, the P2X family of ligand-gated ion channels, and the P2Y family of metabotropic G-protein coupled receptors. To date, seven P2X receptor subtypes and eight different P2Y receptor subtypes have been identified. The P2X receptor subunits exhibit approximately 35%-48% identity with each other at the amino acid level, and have been shown to form both homo- and hetereromultimeric complexes. The various P2X receptor subunits exhibit a broad pattern of distribution, being localized to different types of immune cells, glands, smooth muscle, skeletal muscle, and the central and peripheral nervous systems. The P2X4 receptor subunit is predominantly expressed in the central nervous system and has a widespread distribution as determined byin situ hybridization and immunohistochemistry.
