Poly(ADP-ribose) polymerase (PARP) is a highly conserved nuclear enzyme present in higher eukaryotes. This gene is a member of a family of enzymes that synthesize and transfer ADP-ribose polymers to protein to modify their functional properties. A key role of PARP in the cell is mediating the repair of single-strand DNA nicks. PARP-2 forms homodimers and hetero dimers with PARP-1 in the base excision repair (BER) pathway. To mediate repair PARP interacts with DNA and recruits DNA repair protein XRCC1, DNA ligase III, DNA polymerase beta and a kinase to the site of damage. As a marker for apoptosis, it has been shown that PARP cleavage mediated by caspase-3 activity occurs at the onset of apoptosis, and inhibition of PARP cleavage attenuates apoptosis in vitro.
