Neurofilaments can be defined as the intermediate of 10 nm diameter filaments found in neuronal cells. They are composed of a mixture of subunits which often includes the neurofilament triplet proteins, NF-L, NF-M and NF-H. Neurofilaments may also include peripherin, alpha-internexin, nestin and in some cases vimentin. Antibodies to the variouse neurofilament subunits are very useful cell type markers since the proteins are quite abundant, biochemically stable. Peripherin is a class III intermediate filament subunit protein found in certain classes of neuron, most of which are located in the peripheral nervous system. Certain sensory neurons in the brain stem express peripherin normally, as do a few cortical and hoppocampal neurons. Interestingly, peripherin protein is upregulated in the lower motor neurons that degerate in Amyotrophic Lateral Sclerosis (ALS), focusing attention on peripherin as a potential contributary factor to this serious human neurodegenerative disease. Upregulation on peripherin mRNA and protein expression is seen in a variety of damage and disease states.
