The vascular endothelial growth factor receptor-2 (VEGFR-2,~230kDa) protein is a member of a subfamily of receptor-tyrosine kinases characterized by the presence of seven immunoglobulin-like domains in the extracellular portion of the molecules and a split tyrosine-kinase domain intracellularly. Also known as KDR/Flk-1, VEGFR-2 is expressed primarily in endothelial cells hematopoietic stem cells, megakaryocytes, and retinal progenitor cells. The receptor is a glycoprotein but glycosylation is not required for activity. Unlike the closely related protein, VEGFR-1, VEGFR-2 is only bound by VEGF and not PIGF. Following ligand binding VEGFR-2 undergoes autophosphorylation with the subsequent signal transduction cascades initiated. VEGFR-2 activity is modulated by VEGFR-1 and the two proteins may actually form heterodimers. Interestingly, the neuropilin-1 protein, which has also been shown to be capable of binding to VEGF, may function as a co-receptor for VEGF165 with VEGFR-2.
