The Sec61 complex is responsible for translocation of proteins across the rough endoplasmic reticulum membrane. Sec61 is composed of three subunits: alpha, beta, and gamma; that associate to form a heterotrimer. Multiple heterotrimers associate forming the ER translocon. Sec61 alpha is a polytopic protein that has ten membrane-spanning loops. Sec61 has been found to be tightly associated with membrane bound ribosomes, either directly or through adaptor proteins, and is required for assembly of membrane and secretory proteins.PA3-014 detects human, canine and porcine, rat and mouse sec61 alpha from microsomes and shows no cross-reactivity with other sec61 isoforms.PA3-014 has been successfully used in Western blot procedures. By Western blot, this antibody detects an ~38kDa protein representing sec61 alpha from pig microsomes. The PA3-014 immunogen is a synthetic peptide corresponding to residues C(13) V I L P E I Q K P E R K I Q F K E K(31) of canine sec61 alpha. This sequence is completely conserved between human, mouse, rat, and canine.
