CD107b (LAMP-2) Mouse anti-Human, eFluor 450, Clone: eBioH4B4 (H4B4), eBioscience
Mouse Monoclonal Antibody
Manufacturer: eBioscience 48107841
Description
Description: The eBioH4B4 monoclonal antibody reacts with human CD107b, also known as lysosomal-associated membrane protein-2 (LAMP-2). CD107b is a highly glycosylated, type I transmembrane protein of approximately 105 kDa. It is expressed intracellularly in lysosomal/endosomal membranes in nearly all cells. It is also expressed on the surface of degranulating T cells (to a lesser extent than CD107a) and activated platelets as well as some cancer cells. In humans, mutations in CD107b results in a lysosomal glycogen storage disorder, known as Danon disease. Applications Reported: This eBioH4B4 antibody has been reported for use in flow cytometric analysis. Applications Tested: This eBioH4B4 antibody has been pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product No. 88-8824-00) and protocol. Please refer to "Staining Intracellular Antigens for Flow Cytometry, Protocol A: Two step protocol for intracellular (cytoplasmic) proteins." This may be used at 5 µL (0.25 µg) per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. eFluor 450 is an alternative to Pacific Blue. eFluor 450 emits at 446 nm and is excited with the violet laser line (405 nm).
Please make sure that your instrument is capable of detecting this fluorochrome. Excitation: 405 nm; Emission: 445 nm; Laser: Violet Laser Lysosomes are membrane-bound vacuoles which play a critical role in cellular metabolism. These organelles participate in endocytosis, the synthesis and assembly of acid hydrolases, act as sites for digestion of foreign materials and for specialized autolytic cellular processes. Membrane-associated glycoproteins have an important role in normal lysosomal function. Prior to posttranslational modification, Lysosome Associated Membrane Protein 2 (LAMP 2) is an ~45 kDa polypeptide. Mature, functional LAMP 2 is extensively glycosylated with a variety of different N-linked and O-linked oligosaccharides with a total molecular weight of ~100-110 kDa. LAMP 2 has a structure comprised of a large amino-terminal intra-lysosomal domain, a hydrophobic trans-membrane domain, and a short carboxyl-terminal cytoplasmic tail.Specifications
