Description: The eBioABL-93 monoclonal antibody reacts with mouse CD107b, also known as lysosomal-associated membrane protein-2 (LAMP-2). CD107b is a highly glycosylated protein of approximately 120 kDa that is expressed intracellularly in lysosomal membranes. It is also transiently expressed on the surface of cytolytic T cells during degranulation, but to a lesser extent than CD107a. It has been reported that CD107b may be identical to the mouse Mac-3 antigen. Applications Reported: This eBioABL-93 (ABL-93) antibody has been reported for use in intracellular staining followed by flow cytometric analysis. It is also reported for use in surface staining in a flow cytometric based degranulation assay. Applications Tested: This eBioABL-93 (ABL-93) antibody has been tested by intracellular staining and flow cytometric analysis of thioglycollate elicited peritoneal macrophages. This can be used at less than or equal to 1 µg per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest. Excitation: 488 nm; Emission: 519 nm; Laser: Blue Laser. Filtration: 0.2 µm post-manufacturing filtered.

Lysosomes are membrane-bound vacuoles which play a critical role in cellular metabolism. These organelles participate in endocytosis, the synthesis and assembly of acid hydrolases, act as sites for digestion of foreign materials and for specialized autolytic cellular processes. Membrane-associated glycoproteins have an important role in normal lysosomal function. Prior to posttranslational modification, Lysosome Associated Membrane Protein 2 (LAMP 2) is an ~45 kDa polypeptide. Mature, functional LAMP 2 is extensively glycosylated with a variety of different N-linked and O-linked oligosaccharides with a total molecular weight of ~100-110 kDa. LAMP 2 has a structure comprised of a large amino-terminal intra-lysosomal domain, a hydrophobic trans-membrane domain, and a short carboxyl-terminal cytoplasmic tail.
