Description: The M1/70 monoclonal antibody reacts with mouse CD11b, the 165-170 kDa integrin alphaM. CD11b non-covalently associates with CD18 to form alphaMbeta2 integrin (Mac-1) and binds to CD54 (ICAM-1), C3bi, and fibrinogen. Mac-1 is expressed by macrophages, NK cells, granulocytes, activated lymphocytes and mouse B-1 cells in the peritoneal cavity. M1/70 is also cross-reactive to human CD11b, and can be used for the detection of this antigen on human peripheral blood monocytes, granulocytes, and a subset of NK cells. Through interactions with its ligands, CD11b participates in adhesive cell interactions. Applications Reported: This M1/70 antibody has been reported for use in flow cytometric analysis. Applications Tested: This M1/70 antibody has been tested by flow cytometric analysis of mouse splenocyte or bone marrow cell suspensions. This can be used at less than or equal to 0.25 μg per test. A test is defined as the amount (μg) of antibody that will stain a cell sample in a final volume of 100 μL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest. Super Bright 702 is a tandem dye that can be excited with the violet laser line (405 nm) and emits at 702 nm. We recommend using a 710/50 bandpass filter. Please make sure that your instrument is capable of detecting this fluorochrome.SB-4401) to minimize any non-specific polymer interactions. Please refer to the datasheet for Super Bright Staining Buffer for more information. Light sensitivity: This tandem dye is sensitive to photo-induced oxidation. Please protect this vial and stained samples from light. Fixation: Samples can be stored in IC Fixation Buffer (Product No. 00-8222) (100 μL of cell sample + 100 μL of IC Fixation Buffer) or 1-step Fix/Lyse Solution (Product No. 00-5333) for up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency/compensation. Some generalizations regarding fluorophore performance after fixation can be made, but clone specific performance should be determined empirically. CD11a (LFA-1 alpha, ITGAL) together with CD18 constitute leukocyte function-associated antigen 1 (LFA-1), the alphaLbeta2 integrin. CD11a is implicated in activation of LFA-1 complex. CD11a plays a central role in leukocyte intercellular adhesion through interactions with its ligands, ICAMs 1-3 (intercellular adhesion molecules 1 through 3), and also functions in lymphocyte costimulatory signaling. CD11a is expressed on the plasma membrane of leukocytes in a low-affinity conformation. Cell stimulation by chemokines or other signals leads to induction the high-affinity conformation, which supports tight binding of CD11a to its ligands, the intercellular adhesion molecules ICAM-1, -2, -3. CD11a is thus involved in interaction of various immune cells and in their tissue-specific settlement, but participates also in control of cell differentiation and proliferation and of T-cell effector functions. Blocking of CD11a function by specific antibodies or small molecules has become an important therapeutic approach in treatment of multiple inflammatory diseases. For example, humanized anti- CD11a antibody Efalizumab (Raptiva) is being used to interfere with T cell migration to sites of inflammation, binding of cholesterol-lowering drug simvastatin to CD11a allosteric site leads to immunomodulation and increase in lymphocytic cholinergic activity. Two transcript variants encoding different isoforms of CD11a have been found. Diseases associated with CD11a dysfunction include benign cephalic histiocytosis and leukocyte adhesion deficiency.
