Each compound is created using combinatorial chemistry to form trimers with over 200 building blocks at each of three positions. Each building block is tagged with a unique DNA barcode. When the building blocks are connected through amide bonds, the DNA is also combined forming a double-stranded DNA barcode for each compound. The compound is attached through an amide bond to a C6 linker which is then attached to an oxygen in the 5' phosphate on the DNA.All the compounds fit in a single microcentrifuge tube. The compounds can be incubated with a target of interest that is attached to beads. After washing, the compounds bound to the target can be detected. The DNA barcodes are amplified using PCR and then detected using nextgen sequencing. The sequencing data can be loaded to a secure portal on our website for analysis. This will generate a report of the highest affinity compounds that were bound to the target.Several methods are used to QC the library. The initial single building blocks with their attached DNA barcodes are tested by LC-MS. PCR is used to monitor library assembly, and at completion the library is tested by nextgen sequencing to ensure reasonable compound distribution. The cost of the library includes generating a list of 50 hits or up to 60 hits if there are ties. All the hit structures will be revealed at no additional charge. There are also no additional licensing fees for using the compounds for research and development or manufacturing.
