Detect free calcium ion which serves as a key second messenger in intracellular signaling pathways using Thermo Scientific™ Fluo-8 and Rhod-4 Reagents.

Intracellular calcium levels can be imaged and quantified for numerous biological investigations. Fluorescent probes that show spectral responses upon binding Ca2+ have enabled researchers to investigate changes in intracellular free Ca2+ concentrations by using fluorescence microscopy, flow cytometry, fluorescence spectroscopy and fluorescence microplate readers. Cellomics Fluo-8 and Rhod-4 calcium detection reagents are the brightest green and red calcium indicators. Once inside the cell, the lipophilic blocking groups of Fluo-8 and Rhod-4 are cleaved by esterases, resulting in a negatively charged fluorescent dye that stays inside cells; and its fluorescence is greatly enhanced upon binding to calcium. When cells are stimulated with agonists, the receptor signals the release of intracellular calcium, greatly increasing the fluorescence of Fluo-8 and Rhod-4.

Highlights:
  • Long wavelength, high sensitivity, and fluorescence enhancement (>100 times for Fluo-8 and >250 times for Rhod-4 when it forms a complex with calcium) make these ideal indicators for measurement of intracellular calcium
  • Reagents can be also used to monitor G protein-coupled receptor and calcium channel activities in different cell types
  • Calcium imaging can be performed in a convenient 96-well or 384-well microplate format and is easily adapted to automation in different high-content imaging systems
