Description: The C8.6 antibody reacts with the p40 subunit of human IL-12 p70 and human IL-23. IL-12 is a 70 kDa heterodimeric cytokine composed of two covalently linked chains, p40 and p35, and secreted primarily by activated monocytes, macrophages, and dendritic cells. IL-23 is a biologically-related, heterodimeric cytokine composed of disulfide-linked p40 and p19 subunits. Applications Reported: The C8.6 antibody has been reported for use in ELISA. Applications Tested: The biotinylated C8.6 antibody has been tested as the detection antibody in a sandwich ELISA for analysis of human IL-12 p70 in combination with the purified B-T21 (13-7128) antibody for capture and recombit human IL-12 p70 (14-8129) as the standard. A suitable range of concentrations of this antibody for ELISA detection is 0.5 - 2.0 µg/mL. A standard curve consisting of doubling dilutions of the recombit standard over the range of 2000 pg/mL - 15 pg/mL should be included in each ELISA plate.The biotinylated C8.6 antibody has been tested as the detection antibody in a sandwich ELISA for analysis of human IL-12 p40 monomer in combination with the purified C8.3 (14-7127) antibody for capture and recombit human IL-12 p40 as the standard. A suitable range of concentrations of this antibody for ELISA detection is 0.5-2.0 µg/mL.

A standard curve consisting of doubling dilutions of the recombit standard over the range of 10000 pg/mL - 80 pg/mL should be included in each ELISA plate. Filtration: 0.2 µm post-manufacturing filtered. IL-12 is an interleukin produced by dendritic cells, neutrophils, macrophages and human lymphoblastoid cells as a result of antigenic stimulation. IL-12 is a heterodimeric cytokine encoded by IL-12A (p35) and IL-12B (p40). IL-23 is a heterodimeric cytokine that shares a subunit with IL-12 (IL-12B) along with an IL-23A (IL-23p19) subunit. Functionally, IL-12 increases the expression of vascular endothelial adhesion molecule-1 on the endothelium, directs leukocytes towards tumors and facilitates the ischemic-hemorrhagic necrosis of the tumor tissue. In comparison, IL-23 plays an important role in stimulating memory T-cells, and is required for the induction, expansion, maintece and downstream effector functions of Th17 cells, which play a vital role in upregulating neutrophil chemokines and various pro-inflammatory cytokines.
