The Human Monocyte Chemoattractant Protein-4/CCL1 (Hu MCP-4) ELISA quantitates Hu MCP-4 in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu MCP-4. Principle of the method The Human MCP-4 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Rigorous validation Each manufactured lot of this ELISA kit is quality tested for criteria such as sensitivity, specificity, precision, and lot-to-lot consistency. Cytokines are a family of secreted proteins involved in immunoregulatory and inflammatory processes. The gene encoding MCP-4/CCL13 is one of several Cys-Cys (CC) cytokine genes clustered on the q-arm of chromosome 17. The CC cytokines are proteins characterized by two adjacent cysteines. The cytokine encoded by this gene displays chemotactic activity for monocytes, lymphocytes, basophils and eosinophils, but not neutrophils. MCP-4/CCL13 signals through CCR2B and CCR3 receptors, and plays a role in the accumulation of leukocytes at both sides of allergic and non-allergic inflammation. It be also involved in the recruitment of monocytes into the arterial wall during the disease process of atherosclerosis, and may also play a role in monocyte attraction in tissues chronically exposed to exogenous pathogens.
