Selectively isolate and cultivate Listeria monocytogenes with Thermo Scientific™ Remel™ Fraser Broth.
L. monocytogenes is a common contaminant in raw milk, meat, vegetable, seafood and in food processing environment1,2. Low numbers of L. monocytogenes is improved by enrichment procedures using a broth medium. Fraser and Sperber developed a rapid screening procedure for Listeria species by modifying the U.S. Department of Agriculture (USDA) secondary enrichment broth which improved to selectively isolate and presumptively identify based on esculin hydrolysis3,4.
Fraser Broth is a liquid medium recommended for cultivation of Listeria monocytogenes. This broth improves successful recovery of Listeria species from food and environmental sources. It promotes growth of potentially injured cells and inhibits the growth of non- Listeria background organisms5.
- Ready-to-use: Convenience of prepared media.
- Easy-to-identify: Listeria hydrolyzes esculin to form 6,7-dihydroxycoumarin, which combines with ferric ammonium citrate to blacken the medium.
Fraser Broth contains casein peptone, meat peptone and beef extract that supply amino acids, peptides and nitrogenous subtances essential for of bacteria.Yeast extract supplies essential B-complex vitamins that stimulate growth. Sodium chloride provides essential electrolytes which maintain osmotic equilibrium. Disodium phosphate and monopotassium phosphate are buffers that are added to maintain the pH. Lithium chloride, nalidixic acid, and acriflavine hydrochloride are selective agents. Fraser Broth Additive is added to prepared Fraser Broth base immediately before use, which supplies ferric ammonium citrate.
