AdipoGen BCECF-AM
Chemical. CAS 117464-70-7. Formula C42H40O21. MW 808.69. Synthetic. Widely-used fluorescent indicator for estimating intracellular pH. This membrane-permeant compound is modified by intracellular esterases to produce BCECF, a polar fluorescein derivative that is retained by cells. Intracellular BCECF is stable and has an efflux half-life of greater than two hours. Intracellular pH measurements are made by ratio imaging, which involves determining the pH-dependent ratio of emission intensity detected at 535nm when the dye is excited at 490nm vs. the emission intensity when excited at 440nm. This approach is amenable to either spectrofluorometry or flow cytometry. BCECF-AM ester can also be used to investigate intracellular changes in other ions, including potassium. Spectral properties lambdaex 490nm lambdaem 535nm lambdaex=505nm lambdaem=520nm in DMSO.

AdipoGen 5-ROX

It is preferred for some complicated biological applications where reproducibility is really critical. The minor positional difference between 5-ROX and 6-ROX might significantly affect some biological properties of the underlying conjugates. Used for oligonucleotide labeling and automated DNA sequencing applications.

The amine-reactive fluorescein derivatives are probably the most common fluorescent derivatization reagents for covalently labeling proteins. Carboxyfluorescein is better retained in cells than is fluorescein, its pKa of ~6.5 is lower than the cytosolic pH of most cells pH ~6.8-7.4. Carboxyfluorescein is commonly employed as a polar tracer.

AdipoGen DAF-FM Solution
Chemical. CAS 254109-20-1. Formula C21H14F2N2O5. MW 412.34. Synthetic. DAF-FM is an important reagent for quantitating low concentrations of nitric oxide in solution. This compound is essentially nonfluorescent until it reacts with NO to form a fluorescent benzotrizole. With excitation/emission maxima of 495/515 nm and a dectection limit of 3 nM, DAF-FM can be detected using any instrument that can be used to detect fluorescein. It is more photo-stable and less pH sensitive than DAF-2.
