XL-3158 is a selective and cross-species Cyclic GMP-AMP synthase (cGAS) inhibitor. It simultaneously occupies allosteric and orthosteric sites, stabilizing the activation loop in a closed, inactive conformation, thereby attenuating cGAS-DNA interactions.
