Glycerol Nitrate Agar 25% (G25 N) is a chemically defined selective medium used for the isolation and enumeration of xerophilic fungi that thrive in low moisture environments. Each sterile petri dish contains approximately 30mL of Czapek Dox agar base supplemented with 25% glycerol. The high glycerol concentration significantly reduces water activity, creating conditions that inhibit the growth of many common bacteria and rapidly growing molds. This selective environment allows xerotolerant species such as Aspergillus and Penicillium to grow while suppressing organisms that require higher moisture levels. Nitrate serves as the primary nitrogen source within the medium, supporting fungal metabolic processes. The formulation is widely used for microbiological examination of dried foods, grains, spices, flour, and other low water activity materials where xerophilic fungi are commonly encountered.
