PARP2 (Poly ADP-ribose Polymerase 2) is a DNA repair enzyme in the PARP family that plays an important role in repairing DNA single-strand breaks and maintaining genome stability. PARP2 detects DNA damage, becomes activated, and uses NAD⁺ to generate poly (ADP-ribose) chains through a process called PARylation, which recruits additional DNA repair proteins to damaged sites. PARP2 is overexpressed in several cancers and contributes to tumor cell survival. Certain PARP inhibitors not only block PARP2 catalytic activity but also trap PARP2 on damaged DNA, preventing DNA repair and leading to accumulation of toxic DNA-protein complexes. This effect is especially important in BRCA1/2-mutant cancers.
