NEOGEN Urea Agar Base (Christensen), 500 g
Urea Agar Base is used with agar for the differentiation of microorganisms on the basis of urease production in a laboratory setting. Urea Agar Base is not intended for use in the diagnosis of disease or other conditions in humans. Christensen devised a urea agar medium containing peptone and dextrose that had a reduced buffer content. The medium supported the vigorous growth of many Gram-negative, enteric bacilli and readily permitted observation of urease production. Urea Agar Base may be used as a screening medium (along with Triple Sugar Iron Agar) for the selection of Salmonella and Shigella cultures for serologic classification. Urea Agar Base is also used to detect production of urease by yeast. Dextrose Tryptone Broth is used for cultivation of mesophilic or thermophilic spoilage microorganisms from food in a laboratory setting. Dextrose Tryptone Broth is not intended for use in the diagnosis of disease or other conditions in humans.In the 1930’s, the National Canners Association specified use of Dextrose Tryptone Agar for isolating “flat sour” organisms from food products. “Flat sour” spoilage of canned foods is caused by Bacillus coagulans (Bacillus thermoacidurans). Bacterial growth results in a 0.3 – 0.5 drop in pH, while ends of the can remain flat. B. coagulans is a soil microorganism and found in canned tomato products and dairy products. Due to the high carbohydrate content and low pH this medium is highly sensitive to overheating. NEOGEN Eosin Methylene Blue (EMB) Agar Levine, 5 kg
Eosin Methylene Blue Agar Levine is used for the isolation and differentiation of Gram-negative enteric bacilli and is not intended for use in the diagnosis of disease or other conditions in humans.This medium was introduced in 1916 by Holt-Harris and Teague to differentiate Escherichia spp. and Aerobacter spp. It was modified by Levine in 1918 who removed sucrose from the formula and increased the lactose content. The distinctive metallic sheen produced by E. coli on this medium is due to acid production resulting in an amide bonding between the eosin and methylene blue, other coliforms do not produce enough acid to cause this reaction. This medium is supplemented with oxytetracycline as a selective agent, rather than relying on the low pH of acidified agar to suppress bacterial growth. Casman reported that a medium consisting of 2% Tryptose, along with other media ingredients, equaled fresh beef infusion base with respect to organism growth. The Association of Official Analytical Chemists (AOAC) specifies m-FC Agar for detecting total coliforms and fecal coliforms in foods.
