Rochaix first noted the value of esculin hydrolysis in the identification of enterococcci. Meyer and Schonfeld added bile to the esculin medium and demonstrated 61 of 62 enterococci strains were able to grow and hydrolyze esculin, while other streptococci could not.Molecular taxonomic studies of the genus Streptococcus have placed enterococci, previously described as group D streptococci, in the genus Enterococcus. Skim Milk, Agglomerated NFDM is dehydrated skim milk for use in preparing microbiological culture media in a laboratory setting. Palcam Agar was developed by Van Netten et al in 1989 as an improved selective differential medium for the isolation of Listeria monocytogenes from food and environmental specimens. Improved selectivity is achieved by the combination of antibiotic supplements and microaerobic incubation. The double indicator system of esculin hydrolysis and mannitol fermentation aids differentiation of Listeria spp from enterococci and staphylococci which can be confused with Listeria spp on other types of culture media. Tryptose is an enzymatic digest of protein for use in preparing microbiological culture media in a laboratory setting. Tryptose was developed while studying growth requirements of Brucella spp. Huddleson found Tryptose media to be equal or superior to meat infusion media, providing uniformity for the cultivation and differentiation of fastidious organisms including Neisseria gonorrhoeae, Streptococcus milleri and Brucella spp.. Casman reported that a medium consisting of 2% Tryptose, along with other media ingredients, equaled fresh beef infusion base with respect to organism growth. Tryptose is recommended in broth and agar formulations, and when used in media containing blood to permit hemolytic reactions. Terrific Broth, Modified was developed by Tartoff and Hobbs to increase yields of plasmid DNA from transformed E. coli and is a popular choice of media available for plasmid preps. While cells grown in Terrific Broth, Modified can grow slower than other liquid media, the final cell density and plasmid yields are 2 – 4 fold higher. The higher yield is a result of the increased concentration of Enzymatic Digest of Casein and Yeast Extract in this formula. NEOGEN Lysine Iron Agar, 500 g
Lysine Iron Agar is used for the differentiation of microorganisms on the basis of lysine decarboxylase and hydrogen sulfide production. Lysine Iron Agar is not intended for use in the diagnosis of disease or other conditions in humans. Lysine Iron Agar is prepared according to the formulation of Edwards and Fife, who developed the medium to detect Salmonella arizonae. S. arizonae ferments lactose rapidly, and the authors found expected H2S production on Triple Sugar Iron Agar was suppressed. Detection of S. arizonae is important because it has been implicated in food borne infections. By eliminating lactose and incorporating lysine, Edwards and Fife devised a medium differentiating enteric bacilli based on their ability to decarboxylate or deaminate lysine and produce abundant hydrogen sulfide.
