This cell line enables assessment of chaperone-based stabilization of MC4R(T162I) via detection of rescued membrane-endosome trafficking. The included cell line overexpresses ProLink-tagged mutant MC4R(T162I), which is misfolded and retained in the ER. The cell line also expresses endosome-localized EA. When a pharmacochaperone stabilizes the mutant MC4R, it is trafficked to the membrane and then internalized to the endosome, producing EFC signal. The cell line is engineered to express the catalytic domain from FGR fused to the cytokine receptor binding domain (CRBD) of JAK2. The cells co-express an Enzyme Acceptor (EA) tagged SH2 domain and a ProLink (PK) tagged Cytokine receptor. The CRBD is designed to bind the CTK catalytic domain to the GHR-PK. Activation of the GHR-PK induces receptor phosphorylation by the FGR catalytic domain. This leads to SH2-EA recruitment and production of EFC signal.
