Biotium Cytokeratin 10 (Suprabasal Epithelial Marker) (rKRT10/1275), CF568 conjugate, 0.1mg/mL
This MAb recognizes a protein of 56.5 kDa, identified as cytokeratin 10 (CK10). CK10 is expressed in all suprabasal layers of the epidermis. In the epidermis, expression of CK10 strictly parallels the extent of differentiation; it is absent in the basal layer, appears in the first suprabasal layers and increases in concentration towards the granular layer. However, CK10 is rarely detected in early stages of vulvar squamous carcinomas (tumors less than 2 cm, clinical stage I) regardless of the tumor grade. In larger and more advanced tumors (greater than 2 cm, clinical stages II and III), CK10 is detected very frequently. Expression of CK10 is related to maturation of malignant keratinocytes, being preferentially detected in more-differentiated parts.Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. This antibody recognizes a transmembrane glycoprotein of 95 kDa, identified as CD18 or integrin -2 (Workshop III). It complexes non-covalently with either L, M, or X integrin (CD11a, b, or c) to form the heterodimers, LFA-1, Mac-1, and p150,95, respectively. LFA-1 is the receptor for three members of the Ig supergene family of proteins, ICAM-1 (CD54), ICAM-2 (CD102), and Mac-1 and p150,95 bind to ICAM-1, fibrinogen, and iC3b. ICAM-3 (CD50). CD18/CD11 heterodimeric molecules are involved with cell/cell and cell/extracellular adhesion in immune and inflammatory responses. This antibody recognizes an intermediate filament protein (IFP) of 55 kDa, which is identified as cytokeratin 7. This MAb is highly specific to cytokeratin 7 and shows no cross-reaction with other IFPs. Cytokeratin 7 is a basic cytokeratin, which is found in most glandular and transitional epithelia but not in the stratified squamous epithelia. Keratin 7 is expressed in the epithelial cells of ovary, lung, and breast but not of colon, prostate, or gastrointestinal tract. This MAb recognizes the 56.5 kDa (CK10); 50 kDa (CK14); 50 kDa (CK15); 48 kDa (CK16); 40 kDa (CK19) keratins of the acidic (Type I or LMW) subfamily. Twenty human keratins are resolved with two-dimensional gel electrophoresis into acidic (pI <5.7) and basic (pI >6.0) subfamilies. The acidic keratins have molecular weights (MW) of 56.5, 55, 51, 50, 50 , 48, 46, 45, and 40 kDa.
