This Mab recognizes Mouse DNGR-1, which is also known as CLEC9A, which is a highly specific marker of the CD8a and the CD103 DC subsets, as well as a receptor able to recognize a preformed signal exposed on necrotic cells. Keratins are a family of intermediate filament proteins that assemble into filaments through forming heterodimers of one type I keratin (keratins 9 to 23) and one type II keratin (keratins 1 to 8). Keratins demonstrate tissue and differentiation specific expression profiles. Keratin 15 is a type I keratin which is expressed only in basal keratinocytes in stratified epithelia and does not appear to have a natural type II expression partner. Keratin 15 is down regulated in activated keratinocytes. This antibody recognizes a protein of 95 kDa, which is identified as villin. It is a major constituent in the microvilli, which compose the brush border of epithelial cells forming absorptive surfaces of the intestinal and renal proximal tubular epithelia. Anti-Villin labels the brush border area in the gastrointestinal mucosal epithelium and urogenital tract. Among neoplasms, villin is predominantly expressed in tumors of colorectal origin. Antibody to villin is useful in identifying malignant cells from primary and metastatic colorectal carcinomas. This antibody recognizes a 170 kDa protein, which is identified as topoisomerase IIα. It shows no cross-reaction with topoisomerase IIβ or topoisomerase I. NKX3. 1 is a prostate specific gene encoding a transcription factor that plays an important role in normal prostate development and carcinogenesis. It is a prostatic tumor suppressor gene located on chromosome 8p21. 2, which frequently undergoes a loss of heterozygosity. NKX3. 1 expression is highly restricted in prostate epithelial cells and therefore can be used as a diagnostic biomarker for prostate cancer and other metastatic lesions of prostatic origin. Furthermore, NKX3. 1 shows better sensitivity than Prostate Specific Antigen (PSA) for identifying metastatic prostatic adenocarcinoma. This suggests that immunohistochemical staining of NKX3. 1, along with other prostate-restricted markers, may be valuable for the definitive determination of prostatic origin in poorly differentiated metastatic carcinomas. Recognizes a protein of 66 kDa-170 kDa, identified as involucrin. In Western blotting of cultured human keratinocytes, this MAb reacts with a 120 kDa protein. It stains the involucrin in a variety of sizes: 170 kDa in MCF-7 cells, a doublet of ~115 kDa and 150 kDa in gorilla and owl monkey, 66 kDa in dog, and a doublet of 105 kDa in pig. Its epitope maps between codon 421-568 of human involucrin. Involucrin is expressed in a range of stratified squamous epithelia, including the cornea, which lacks a distinct cornified layer. This MAb recognizes a protein of 52 kDa, which is identified as TNFR1 (TNFR-associated factor 1). CD30-positive lymphoproliferations of the skin comprise 30% of all primary cutaneous T-cell lymphomas (CTCLs). Besides borderline cases this group includes lymphomatoid papulosis (LyP) and primary cutaneous anaplastic large T-cell lymphoma (cALCL). Although the two entities overlap clinically, histopathologically, immunopathologically and genetically, they differ considerably in their prognosis. In particular, common feature of both cases is histologically the presence of atypical lymphoid CD30-positive T blasts and genetically a clonal T-cell-receptor rearrangement. This antibody recognizes proteins of 80-200 kDa, identified as different members of CEA family. CEA is synthesized during development in the fetal gut and is re-expressed in increased amounts in intestinal carcinomas and several other tumors. This MAb does not react with nonspecific cross-reacting antigen (NCA) and with human polymorphonuclear leucocytes. It shows no reaction with a variety of normal tissues and is suitable for staining of formalin/paraffin tissues. CEA is not found in benign glands, stroma, or malignant prostatic cells. This antibody recognizes a protein of 116 kDa, identified as P-Cadherin-1 (CDH3). It is a calcium-dependent cell-cell adhesion glycoprotein comprised of five extracellular cadherin repeats, a transmembrane region and a highly conserved cytoplasmic tail. This gene is located in a six-cadherin cluster in a region on the long arm of chromosome 16 that is involved in loss of heterozygosity events in breast and prostate cancer. Recognizes a 67 kDa glycoprotein, which is identified as CD33 (HLDA IV; WS Code M-505). CD33 is a transmembrane protein of the sialic acid-binding immunoglobulin-like lectin (Siglec) family. It belongs to the immuno-receptor tyrosine-based inhibitory motif (ITIM)-containing molecules able of recruiting protein tyrosine phosphatases SHP-1 and SHP-2 to signal assemblies; these ITIMs are also used for ubiquitin-mediated removal of the receptor from the cell surface.
