Complement C1q A Chain (C1qa) is a subunit of C1q. C1q is composed of 6 polypeptide chains each of C1qa, C1qb, and C1qc. C1q is a component of the classical complement pathway, and thereby plays an important role in the immune response. Each C1q subunit contains an N-terminal collagen-like region and a C-terminal C1q globular domain. The presence of receptors for C1q on effector cells modulates its activity, which may be antibody-dependent or independent. Macrophages are the primary source of C1q, while anti-inflammatory drugs as well as cytokines differentially regulate expression of the mRNA as well as the protein. Biotium MUC2 (Mucin 2) (MLP/2970R), CF647 conjugate, 0.1mg/mL
This antibody recognizes a single glycoprotein of 520 kDa, identified as mucin 2 (MUC2). This MAb shows no cross-reaction with human milk fat globule membranes, MUC1, or MUC3. Its epitope has been defined as GTQTP (GlyThrGlnThrPro). Mucins are high molecular weight glycoproteins, which constitute the major component of the mucus layer that protects the gastric epithelium. MUC2 is specifically expressed in goblet cells of the small intestine & colon; in about 65% of colonic carcinomas and about 40% of gastric carcinomas. Recognizes a single protein of 33-34 kDa, identified as the prostate specific antigen (PSA). This MAb is highly specific to PSA and stains prostatic secretory and ductal epithelium in both normal and neoplastic tissues. PSA is a chymotrypsin-like serine protease (kallikrein family) exclusively produced by the prostate epithelium, and abundant in seminal fluid. PSA can be detected in the sera of patients with prostatic carcinoma. It is predominantly complexed to a liver-derived serine protease inhibitor, alpha-1-antichymotrypsin (ACT). A higher proportion of serum PSA is complexed to ACT in prostate cancer than in benign prostate hyperplasia.

CD9 is a type IV transmembrane glycoprotein with four transmembrane domains. CD9 on pre-B cells may play a role in cell-cell adhesion. In addition, CD9 may play a role in signal transduction mediated by interaction with low molecular weight GTP binding proteins. CD9 is expressed on early B cells, eosinophils, basophils and activated T cells and is a major component of the platelet cell surface. Biotium Ksp-Cadherin (Kidney-Specific Cadherin) / CDH16, CF647 conjugate, 0.1mg/mL
This antibody recognizes a protein of 130 kDa, identified as Ksp-cadherin. Cadherins form a superfamily of related glycoproteins that mediate calcium-dependent cell adhesion and transmit signals from the extracellular matrix to the cytoplasm. Cadherins have been implicated in embryogenesis, tissue morphogenesis, tissue structure maintenance, cell polarization, neoplastic invasiveness and metastasis, and membrane transport. It is suggested that Ksp-cadherin is a marker for terminal differentiation of the basolateral membranes of renal tubular epithelial cells. Within the kidney, Ksp-Cadherin is found exclusively in the basolateral membrane of renal tubular epithelial cells and collecting duct cells, and not in glomeruli, renal interstitial cells, or blood vessels. The progression of cells through the cell cycle is regulated by a family of proteins designated cyclin-dependent kinases (Cdks). Sequential activation of individual members of this family and their consequent phosphorylation of critical substrates promotes orderly progression through the cell cycle. Multiple proteins are encoded by the tumor suppressor gene CDKN2A (MTS1/ p16INK4a) via translation of alternate reading frames, resulting in the pro- duction of the p19 ARF protein in mice and the p14 ARF protein in humans. p14 ARF induces an increase in MDM2 and p21 levels and leads to cell cycle arrest in both G1 and G2/M. p14 ARF is negatively regulated by p53 and is known to bind directly to MDM2. CDKN2A also encodes the mitotic protein p16, which binds to and inhibits the Cdk4/cyclin D complex. This antibody recognizes a protein of ~35 kDa, identified as CD74 (Workshop IV). CD74 is a type II transmembrane protein which binds to the peptide binding groove of newly synthesized MHC class II alpha/beta heterodimers and prevents their premature association with endogenous polypeptides. CD74 is expressed primarily by antigen presenting cells, such as B-lymphocytes (from before the pre-B cell stage to before the plasma cell stage), macrophages, and monocytes, and many epithelial cells. Anti-CD74 stains predominantly germinal center lymphocytes and B-cell lymphomas, but rarely T-cell lymphomas. This antibody recognizes a protein of about 50 kDa, which is identified as gamma-enolase. Three isoenzymes of enolases are identified, alpha, beta and gamma. Alpha-isoform is expressed in most tissues, whereas beta-form is expressed predominantly in muscle tissue whereas gamma-enolase is found only in nervous tissue. These isoforms exist as both homodimers and heterodimers, and they play a role in converting phosphoglyceric acid to phosphenolpyruvic acid in the glycolytic pathway. NSE-gamma is a useful marker to identify peripheral nerves and tumors of neuro-endocrine origins, such as pheochromocytomas. The finding that mutations in DNA mismatch repair genes are associated with hereditary nonpolyposis colorectal cancer (HNPCC) has resulted in considerable interest in the understanding of the mechanism of DNA mismatch repair. Initially, inherited mutations in the MSH2 and MLH1 homologs of the bacterial DNA mismatch repair genes mutS and mutL were demonstrated at high frequency in HNPCC and were shown to be associated with microsatellite instability. A member of the mismatch repair family, GTBP (also designated MSH6), is an MSH2-related protein that binds to DNA containing G/T mismatches. Findings suggest that the mismatch-binding factor in human cells is composed of a heterodimer of GTBP and MSH2.
