The C-type lectin superfamily member CD72 is a cell surface negative regulator of B cell activation from the pro-B through the mature B cell stage. CD72 serves as a receptor for CD5. The human CD72 gene encodes a transmembrane glycoprotein that contains an immunoreceptor tyrosine-based inhibition motif (ITIM). Upon tyrosine phosphorylation, the CD72 ITIM recruits SH2-containing phosphatases such as SHP-1, resulting in downregulation of cell activation. CD72-/- mice contain hyperproliferative B cells. This gene encodes a receptor that binds the Fc region of monomeric immunoglobulin G. FCGRT is present in the intestinal epithelium of neonates and mediates the selective uptake of immunoglobulin G (IgG) in mothers' milk, thereby helping newborn to acquire passive immunity. FCGRT is comprised of a heavy chain and -2-Microglobulin. FCGRT localizes in endosomes of vascular endothelial cells and selectively recycles IgG to the cell surface, thus protecting IgG from lysosomal degradation. CD8 is a cell surface receptor expressed either as a heterodimer with the CD8 beta chain (CD8 alpha/beta) or as a homodimer (CD8 alpha/alpha). A majority of thymocytes and a subpopulation of mature T cells and NK cells express CD8a. CD8 binds to MHC class 1 and through its association with protein tyrosine kinase p56lck plays a role in T cell development and activation of mature T cells. For mature T-cells, CD4 and CD8 are mutually exclusive, so anti-CD8, generally used in conjunction with anti-CD4. It is a useful marker for distinguishing helper/inducer T-lymphocytes, and most peripheral T-cell lymphomas are CD4 /CD8-. Anaplastic large cell lymphoma is usually CD4 and CD8-, and in T-lymphoblastic lymphoma/leukemia, CD4 and CD8 are often co-expressed. CD8 is also found in littoral cell angioma of the spleen. Recognizes a 180-185 kDa protein, identified as isoform of leukocyte common antigen (CD45RO) (4th Leucocyte Typing Workshop: Code No. N31). The epitope recognized by this antibody is sensitive to neuraminidase digestion. This antibody reacts with mature activated T-cells, most thymocytes, and a sub-population of resting T-cells within both CD4 and CD8 subsets. It shows no reactivity with normal B or natural killer cells, but reacts with granulocytes and monocytes. Reportedly, it is useful to identify T-cell lymphomas and leukemias. S100A4 belongs to the S100 super-family of proteins containing 2 EF-hand calcium-binding domains. S100A4 has been implicated in the progression and prognosis of several forms of human cancer, e. g. breast, colorectal, gastric, pancreatic and bladder cancer, SCLC and oesophageal squamous cell carcinoma, among others. Poor prognosis associated with high S100A4 expression is accompanied by clear signs of disease progression, e. g. high histological and clinical grades and involvement of lymph nodes. Also indicative of poor prognosis is high S100A4 expression coupled with reduced E-cadherin expression in pancreatic, oral squamous cell carcinoma and in melanoma. Geminin is a nuclear protein that regulates the initiation of DNA replication during the cell cycle. DNA replication requires the coordinated association of Cdc6 and minichromosome maintenance (MCM) proteins with chromatin. Geminin blocks this assembly of the MCM into the prereplication complex and, in turn, prevents replication from occurring. Expression of Geminin fluctuates throughout the cell cycle with Geminin levels lowest at G1. This MAb recognizes a protein of 56.5 kDa, identified as cytokeratin 10 (CK10). CK10 is expressed in all suprabasal layers of the epidermis. In the epidermis, expression of CK10 strictly parallels the extent of differentiation; it is absent in the basal layer, appears in the first suprabasal layers and increases in concentration towards the granular layer. However, CK10 is rarely detected in early stages of vulvar squamous carcinomas (tumors less than 2 cm, clinical stage I) regardless of the tumor grade. In larger and more advanced tumors (greater than 2 cm, clinical stages II and III), CK10 is detected very frequently. The epitope of this antibody spans the Myc tag epitope, aa 410-419 (EQKLISEEDL), which is a specific portion of an alpha helical region of human c-myc protein. Because the epitope is located at the dimerization site of the protein, the antibody is not sensitive for detecting endogenous c-myc, making it suitable for detecting Myc tagged proteins or myc overexpression. This MAb shows no cross-reaction with v-myc. c-myc is involved in the control of cell proliferation and differentiation and is amplified and/or overexpressed in a variety of tumors. NKX3. 1 is a prostate specific gene encoding a transcription factor that plays an important role in normal prostate development and carcinogenesis. It is a prostatic tumor suppressor gene located on chromosome 8p21. 2, which frequently undergoes a loss of heterozygosity. NKX3. 1 expression is highly restricted in prostate epithelial cells and therefore can be used as a diagnostic biomarker for prostate cancer and other metastatic lesions of prostatic origin. This antibody recognizes a protein of ~150 kDa, identified as Desmoglein-1 (DSG1). Desmoglein-1 is a member of the desmosomal cadherin family. Desmosomes are intercellular adhering junctions that represent cell surface attachment sites for intermediate filament. Desmocollins and desmogleins are the main desmosomal transmembrane proteins. Desmogleins consist of Dsg1, Dsg2, Dsg3, and Dsg4 isoforms.
