Aldehyde dehydrogenase 1 family member A1 (ALDH1A1), also known as retinal dehydrogenase 1, belongs to the aldehyde dehydrogenase enzyme family, which is involved in the metabolism of alcohol. ALDH1A1 is predominantly expressed in the epithelium of testis, brain, eye, liver, kidney, as well as neural and hematopoietic stem cells. Reportedly, high ALDH1A1 expression is found in solitary fibrous tumor (SFT) and hemangiopericytoma (HPC), compared to meningiomas and synovial sarcomas. In combination with CD34, ALDH1A1 may be useful for the differentiation among SFT, HPC, meningioma, and synovial sarcoma. Expression of NKX2.2 has been found in neuroendocrine tumors of the gut, making it a potential marker for the study of gastrointestinal neuroendocrine tumors. More recently, NKX2.2 protein was identified as a target of EWS-FLI-1, the fusion protein specific to Ewing sarcoma, and was shown to be differentially upregulated in Ewing sarcoma on the basis of array-based gene expression analysis. This MAb recognizes a protein of 56.5 kDa, identified as cytokeratin 10 (CK10). CK10 is expressed in all suprabasal layers of the epidermis. In the epidermis, expression of CK10 strictly parallels the extent of differentiation; it is absent in the basal layer, appears in the first suprabasal layers and increases in concentration towards the granular layer. However, CK10 is rarely detected in early stages of vulvar squamous carcinomas (tumors less than 2 cm, clinical stage I) regardless of the tumor grade. In larger and more advanced tumors (greater than 2 cm, clinical stages II and III), CK10 is detected very frequently. PAI-RBP1 (plasminogen activator inhibitor 1 RNA-binding protein), also known as SERBP1 (SERPINE1 mRNA-binding protein 1), CGI-55, CHD3IP (chromodomain helicase DNA binding protein 3 interacting protein), HABP4L or PAI-RBP1, is a membrane-associated protein that localizes to the nucleus, the perinuclear region of the cytoplasm and the plasma membrane. PAI-RBP1 is believed to play a role in the regulation of mRNA stability, as it specifically binds to the CRS (cyclic nucleotide-responsive sequence) motif of the PAI-1 mRNA and acts to stabilize the mRNA and regulate its expression. In addition, PAI-RBP1 interacts with Mi2- and may be involved in chromatin remodeling. PAI-RBP1 also interacts with PGRMC1 and participates in the transduction of Progesterone's antiapoptotic action in ovarian cell types. Recognizes a protein of 94 kDa, which is identified as the glucose-regulated protein 94 (grp94) and also tumor rejection antigen (gp96). Grp94 shows a high degree of sequence homology with the heat shock protein 90 (hsp90). This MAb is highly specific to grp94 and shows minimal cross-reaction with other members of the HSP90 family. Grp s are a class of proteins unresponsive to heat shock and are induced by glucose deprivation. CD105/Endoglin is a Type I membrane glycoprotein located on cell surfaces and is part of the TGF-beta receptor complex. This protein has been found on endothelial cells, activated macrophages, fibroblasts, and smooth-muscle cells. Endoglin has a role in the development of the cardiovascular system and in vascular remodeling. Its expression is regulated during heart development. CD105 is highly expressed in endothelial cells during tumor angiogenesis and inflammation, with weak or negative expression in vascular endothelium of normal tissues. Angiogenesis is a promising prognostic marker in a variety of tumors. PMEPA1 (prostate transmembrane protein, androgen induced 1 is a 287 amino acid single-pass membrane protein that contains WW-binding motifs and localizes to the cell membrane. Expressed at high levels in prostate, kidney and ovary, PMEPA1 interacts with NEDD4 and may play a role in regulating AR (androgen receptor) levels, specifically in prostate cells. Down regulation of PMEPA1 is observed in prostate tumors, suggesting that PMEPA1 may exhibit activity as a tumor suppressor. Cadherin 17, also known as LI cadherin (for liver-intestine cadherin), is a member of the cadherin family of calcium-dependent adhesion molecules. Cadherins mediate cell-cell interactions via hemophilic binding, and are critical to the maintenance of tissue structure and morphogenesis. Cadherins each contain a large extracellular domain at the amino terminus, which is characterized by a series of five homologous repeats, the most distal of which is thought to be responsible for binding specificity. The relatively short carboxy terminal, intracellular domain interacts with a variety of cytoplasmic proteins, including beta-catenin, to regulate cadherin function. Glypican-3 (GPC3) is a glycosylphospatidyl inositol-anchored membrane protein, which may also be found in secreted form. Anti-GPC3 has been identified as a useful tumor marker for the diagnosis of hepatocellular carcinoma (HCC), hepatoblastoma, melanoma, testicular germ cell tumors, and Wilm s tumor. In patients with HCC, GPC3 is overexpressed in neoplastic liver tissue and elevated in serum, but is undetectable in normal liver, benign liver, and the serum of healthy donors. GPC3 expression is also found to be higher in HCC liver tissue than in cirrhotic liver or liver with focal lesions such as dysplastic nodules and areas of hepatic adenoma (HA) with malignant transformation. This MAb recognizes proteins of 19-22 kDa (reducing) and 38 kDa-44 kDa (non-reducing), identified as various isoforms of Vascular Endothelial Growth Factor or Vascular Permeability Factor (VEGF/VPF). It is highly specific to VEGF, which is a homodimeric, disulfide-linked glycoprotein with a close homology to platelet-derived growth factor (PDGF). There are multiple isoforms of VEGF containing 206-, 189-, 165-, and 121-amino acid residues.
