Calponin is a calmodulin, F-actin and tropomyosin binding protein, which is thought to be involved in the regulation of smooth muscle contraction. Calponin expression is restricted to smooth muscle cells and has been shown to be a marker of the differentiated (contractile) phenotype of developing smooth muscle. Multiple isoelectric variants of calponin have been identified, however only two molecular weight isoforms exist, a 34 kDa form and a 29 kDa form. Expression of the 29 kDa form, I-calponin, is primarily restricted to muscle of the urogenital tract, whereas the higher molecular weight variant has been demonstrated in vascular and visceral smooth muscle. CD50 is the major ligand for LFA-1 (CD11a/CD18) and may have signaling role to increase adhesion. It is expressed on thymocytes and T lymphocytes and is resistant to treatment with phosphatidylinositol (PI) phospholipase C. The LMO2 protein has a central and crucial role in hematopoietic development and is highly conserved. It has a particular function in normal and lymphatic endothelial cells involving the regulation of angiogenesis and lymph-angiogenesis. Immunohistochemical studies have also demonstrated expression of LMO2 in both normal germinal center B-cells and germinal center-derived B-cell lymphomas, including follicular lymphoma and diffuse large B-cell lymphoma. The use of anti-LMO2 is valuable as a tool in the identification of lymphomas of B-cell origin. LMO2 is useful in differentiating follicular lymphoma (LMO2 ) from nodal marginal zone lymphoma (LM02-). It also is positive in Hodgkin's and Burkitt's lymphomas. This antibody recognizes a protein of 75 kDa, identified as mu heavy chain of human immunoglobulins. It does not cross-react with alpha (IgA), gamma (IgG), epsilon (IgE), or delta (IgD), heavy chains, T-cells, monocytes, granulocytes, or erythrocytes. Monomeric IgM is expressed as a membrane bound antibody on the surface of B cells and as a pentamer when secreted by plasma cells. IgM antibody is prominent in early immune responses to most antigens. Aberrant levels are associated with immune deficiency states, hereditary deficiencies, myeloma, Waldenstrom's macroglobulinemia, chronic infection and hepatocellular disease. This MAb is useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. S100A and S100B proteins are two members of the S100 family of calcium binding proteins. S100A is composed of an alpha and a beta chain whereas S100B is composed of two beta chains. This antibody is specific against an epitope located on the alpha-chain (i.e. in S-100A and S-100B) but not on the beta-chain of S-100 (i.e. in S-100B). This antibody can be used to localize S-100A in various tissue sections. S-100 protein has been found in normal melanocytes, Langerhans cells, histiocytes, chondrocytes, lipocytes, skeletal and cardiac muscle, epithelial and myoepithelial cells of the breast, salivary and sweat glands. Neoplasms derived from these cells also express S-100 protein.
