This antibody recognizes a protein of 120-80 kDa, identified as E-cadherin. Cadherins comprise a family of Ca2 -dependent adhesion molecules that function to mediate cell-cell binding critical to the maintenance of tissue structure and morphogenesis. The classical cadherins, E-, N- and P-cadherin, consist of large extracellular domains characterized by a series of five homologous NH2 terminal repeats. The relatively short intracellular domains interact with a variety of cytoplasmic proteins, such as β-catenin, to regulate cadherin function. E-cadherin plays an important role in epithelial cell adhesion. A decreased expression of E-cadherin is associated with metastatic potential and poor prognosis in breast cancer, prostate and esophageal cancer. This antibody recognizes an epitope encoded by exon v9 on the variant portion of human CD44. The CD44 molecule belongs to a family of cellular adhesion molecules found on a wide range of normal and malignant cells in epithelial, mesothelial and hemopoiesis tissues. CD44 is a single gene with 20 exons, of which 10 are normally expressed to encode the basic CD44 (H-CAM) molecule. The additional 10 exons (v1 to v10) are only expressed by alternative splicing of the nuclear RNA. Cytochrome C is a well-characterized mobile electron transport protein that is essential to energy conversion in all aerobic organisms. In mammalian cells, this highly conserved protein is normally localized to the mitochondrial inter-membrane space. More recent studies have identified cytosolic cytochrome c as a factor necessary for activation of apoptosis. During apoptosis, cytochrome c is trans-located from the mitochondrial membrane to the cytosol, where it is required for activation of caspase-3 (CPP32). Overexpression of Bcl-2 has been shown to prevent the translocation of cytochrome c, thereby blocking the apoptotic process. This antibody reacts with a carbohydrate determinant, identified as CDw78 (Workshop V; Code CDw78.2). Epitope recognized by this MAb and Leu21 is the same. It is present on some immature and some peripheral blood B cells, and some B cell lines. Its expression is increased after B-cell activation in vitro. CDw78 is also found on tissue macrophages and on epithelial cells, but not on T cells, NK cells, monocytes, granulocytes and myeloid tissues. This MAb recognizes the peptide core of gastric mucin M1 (recently identified as Mucin 5AC). Its epitope is located in the N-terminal cysteine rich part of the peptide core of MUC5AC, which is heavily glycosylated. Its epitope is destroyed by beta-mercaptoethanol but not by periodate treatment. MAb 2-11M1 reacts with the protein backbone exclusively; it only reacts with fully deglycosylated MUC5AC. Therefore, the material under test should also be fully deglycosylated. This can be achieved with standard periodate oxidation method. The success of the deglycosylation can be checked with routine PAS (Periodic Acid Shiff) staining. After deglycosylation, the preparation should no longer be stainable with PAS reagent. Only then 2-11M1 will react should MUC5AC be present. This antibody recognizes a 32 kDa protein, which is identified as homodimer of galectin-13 (also known as PP13). Recognizes a single protein of 33-34 kDa, identified as the prostate specific antigen (PSA). This MAb is highly specific to PSA and stains prostatic secretory and ductal epithelium in both normal and neoplastic tissues. PSA is a chymotrypsin-like serine protease (kallikrein family) exclusively produced by the prostate epithelium, and abundant in seminal fluid. PSA can be detected in the sera of patients with prostatic carcinoma. It is predominantly complexed to a liver-derived serine protease inhibitor, alpha-1-antichymotrypsin (ACT). A higher proportion of serum PSA is complexed to ACT in prostate cancer than in benign prostate hyperplasia.
