This MAb recognizes a protein of 170 kDa, identified as EGFR. EGFR is type I receptor tyrosine kinase with sequence homology to erbB-1, -2, -3 -4 or HER-1, -2, -3 -4. It binds to Epidermal Growth Factor (EGF), Transforming Growth Factor-a (TGF-a), Heparin-binding EGF (HB-EGF), amphiregulin, βcellulin and epiregulin. EGFR is overexpressed in tumors of breast, brain, bladder, lung, gastric, head & neck, esophagus, cervix, vulva, ovary, and endometrium. This MAb reacts with a 200 kDa and 68 kDa protein, identified as heavy and light sub-units of neurofilaments (NF-H NF-L). Neurofilaments make up the main structural elements of axons and dendrites and are found in neurons, peripheral nerves, and sympathetic ganglion cells. Neurofilaments consist of three major subunits with molecular weights of 68 kDa (NF-L), 160 kDa (NF-M) and 200 kDa (NF-H). Anti-neurofilament stains a number of neural, neuroendocrine, and endocrine tumors. Neuromas, ganglioneuromas, gangliogliomas, ganglioneuroblastomas, and neuroblastomas stain positively for anti-neurofilament. Neurofilaments are also present in paragangliomas as well as adrenal and extra-adrenal pheochromocytomas. Keratin 15 is a type I keratin which is expressed only in basal keratinocytes in stratified epithelia and does not appear to have a natural type II expression partner. Ezrin, Moesin and Radixin belong to a family of highly homologous actin-associated proteins that are localized just beneath the plasma membrane. The proteins are believed to be involved in the mediation of interactions between cytoskeletal and membrane proteins. Ezrin serves as a major cytoplasmic substrate of various protein-tyrosine kinases, including the epidermal growth factor receptor. Ezrin has also been identified as a cAMP-dependent protein kinase (A-kinase) anchoring protein and designated AKAP78. Moesin and Radixin share over 70% homology with Ezrin and are co-expressed within various cell types. This MAb recognizes a protein of 130 kDa, identified as Ksp-cadherin. Cadherins form a superfamily of related glycoproteins that mediate calcium-dependent cell adhesion and transmit signals from the extracellular matrix to the cytoplasm.
