Recognizes a protein of 70 kDa, which is identified as CD86. CD86, along with CD80/B71, is an important accessory molecule in T cell co-stimulation via its interaction with CD28 and CD152/CTLA4, a cancer immunotherapy target. It is a type I transmembrane glycoprotein and a member of the immunoglobulin superfamily of cell surface receptors. It is expressed at high levels on resting peripheral monocytes and dendritic cells and at very low density on resting B and T lymphocytes. CD86 expression is rapidly upregulated by B cell specific stimuli with peak expression at 18 to 42 hours after stimulation. Since CD86 has rapid kinetics of induction, it is believed to be the major CD28 ligand expressed early in the immune response. Reacts with a 75 kDa melanocyte-specific gene product, identified as Tyrosinase-related protein-1 (TRP-1). It is involved in melanin synthesis. TRP1 is present on the melanosomal membranes of melanoma, normal melanocytes and nevi. Recent evidence suggests that TRP-1 is involved in maintaining stability of tyrosinase protein and modulating its catalytic activity. TRP-1 is also involved in maintenance of melanosome ultrastructure and affects melanocyte proliferation and cell death. Filaggrin is an intermediate filament-associated protein that aggregates keratin intermediate filaments in mammalian epidermis. It is initially synthesized as a polyprotein precursor, profilaggrin (consisting of multiple filaggrin units of 324 aa each), which is localized in keratohyalin granules, and is subsequently proteolytically processed into individual functional filaggrin molecules. CD6 is a type I transmembrane glycoprotein that contains a 24-amino acid signal sequence, three extracellular scavenger receptor cysteine-rich (SRCR) domains, a membrane-spanning domain and a 44-amino acid cytoplasmic domain. The CD6 glycoprotein is tyrosine phosphorylated during TCR-mediated T cell activation. CD6 shows significant homology to CD5. CD6 is present on mature thymocytes, peripheral T cells and a subset of B cells. VISTA / GI24 is a transmembrane protein expressed in bone, on embryonic stem cells (ESCs), and on tumor cell surfaces. On ESC s, Gi24 appears to positively interact with BMP-4, potentiating BMP signaling and the transition from an undifferentiated to a differentiated state. On tumor cells, Gi24 both promotes MT1-MMP expression and activity and serves as a substrate for MT1-MMP. This increases the potential for cell motility. This MAb is specific to lambda light chain of immunoglobulin and shows no cross-reaction with lambda light chain or any of the five heavy chains. In mammals, the two light chains in an antibody are always identical, with only one type of light chain, kappa or lambda. The ratio of Kappa to Lambda is 70:30. However, with the occurrence of multiple myeloma or other B-cell malignancies this ratio is disturbed. Recognizes a protein of 57 kDa, identified as p57Kip2. It shows no cross-reaction with p27Kip1. p57Kip2 is a potent tight-binding inhibitor of several G1 cyclin complexes, and is a negative regulator of cell proliferation. Anti-p57 has been used as an aide in identification of complete hydatidiform mole (CHM) (no nuclear labeling of cytotrophoblasts and stromal cells) from partial hydatidiform mole (PHM) in which both cytotrophoblasts and stromal cells stain. The histological differentiation of complete mole, partial mole, and hydropic spontaneous abortion is problematic. This antibody recognizes a carbohydrate epitope on a single chain, transmembrane, heavily glycosylated protein of 90-120 kDa, which is identified as CD34 (VI international workshop on human differentiation antigens). Its expression is a hallmark for identifying pluripotent hematopoietic stem or progenitor cells. Its expression is gradually lost as lineage committed progenitors differentiate. CD34 is a marker of choice for staining blasts in acute myeloid leukemia. In addition, it is expressed by soft tissue tumors, such as solitary fibrous tumor and gastrointestinal stromal tumor. Keratin 15 is a type I keratin which is expressed only in basal keratinocytes in stratified epithelia and does not appear to have a natural type II expression partner. Keratin 15 is down regulated in activated keratinocytes. Cytokeratin 15 is a specific marker of stem cells of the hair-follicle bulge and may be a useful marker for diagnosis between basal cell carcinoma (BCC) and trichoepithelioma. Trichoblastoma are benign neoplasms of follicular differentiation frequently found in nevus sebaceous. Many morphologic features are shared with nodular basal cell carcinoma, sometimes rendering a diagnosis difficult. This antibody recognizes an epitope encoded by exon v3 on the variant portion of human CD44. The CD44 molecule belongs to a family of cellular adhesion molecules found on a wide range of normal and malignant cells in epithelial, mesothelial and hemopoiesis tissues. CD44 is a single gene with 20 exons, of which 10 are normally expressed to encode the basic CD44 (H-CAM) molecule.
