SOX2 is required for stem cell maintenance in the central nervous system, and it also regulates gene expression in the stomach. SOX2 is necessary for regulating multiple transcription factors that affect Oct 3/4 expression. An essential function of SOX2 is to stabilize embryonic stem cells in a pluripotent state by maintaining the requisite level of Oct 3/4 expression. Reportedly, SOX2 is associated with aggressive phenotypes of breast, head and neck, gastric, colorectal, bladder, and small cell lung cancers. However, SOX2 is expressed in a high percentage of lung squamous cell carcinomas and has been shown to be an independent favorable prognostic marker. The epitope of this antibody spans the Myc tag epitope, aa 410-419 (EQKLISEEDL), which is a specific portion of an alpha helical region of human c-myc protein. Because the epitope is located at the dimerization site of the protein, the antibody is not sensitive for detecting endogenous c-myc, making it suitable for detecting Myc tagged proteins or myc overexpression. This antibody recognizes a carbohydrate epitope on a single chain, transmembrane, heavily glycosylated protein of 90-120 kDa, which is identified as CD34 (VI international workshop on human differentiation antigens). Its expression is a hallmark for identifying pluripotent hematopoietic stem or progenitor cells. Its expression is gradually lost as lineage committed progenitors differentiate. CD34 is a marker of choice for staining blasts in acute myeloid leukemia. In addition, it is expressed by soft tissue tumors, such as solitary fibrous tumor and gastrointestinal stromal tumor. CD34 expression is also found in vascular endothelium. Additionally, proliferating endothelial cells overexpress this molecule than the non-proliferating endothelial cells. Myogenin is a member of the MyoD family of myogenic basic helix-loop-helix (bHLH) transcription factors that also includes MyoD, Myf-5, and MRF4 (also known as herculinor Myf-6). MyoD family members are expressed exclusively in skeletal muscle and play a key role in activating myogenesis by binding to enhancer sequences of muscle-specific genes. The regulatory domain of MyoD is approximately 70 amino acids in length and includes both a basic DNA binding motif and a bHLH dimerization motif. MyoD family members share about 80% amino acid homology in their bHLH motifs. Anti-myogenin labels the nuclei of myoblasts in developing muscle tissue, and is expressed in tumor cell nuclei of rhabdomyosarcoma and some leiomyosarcomas.
