This MAb reacts with a protein of 20-30 kDa, identified as PGP9.5, also known as ubiquitin carboxyl-terminal hydrolase-1 (UchL1). Initially, PGP9.5 expression in normal tissues was reported in neurons and neuroendocrine cells but later it was found in distal renal tubular epithelium, spermatogonia, Leydig cells, oocytes, melanocytes, prostatic secretory epithelium, ejaculatory duct cells, epididymis, mammary epithelial cells, Merkel cells, and dermal fibroblasts. Furthermore, immunostaining for PGP9.5 has been shown in a wide variety of mesenchymal neoplasms as well. The epitope of this antibody spans the Myc tag epitope, aa 410-419 (EQKLISEEDL), which is a specific portion of an alpha helical region of human c-myc protein. Because the epitope is located at the dimerization site of the protein, the antibody is not sensitive for detecting endogenous c-myc, making it suitable for detecting Myc tagged proteins or myc overexpression. This MAb shows no cross-reaction with v-myc. c-myc is involved in the control of cell proliferation and differentiation and is amplified and/or overexpressed in a variety of tumors. This antibody recognizes a protein of 21 kDa, identified as the Bax protein. This MAb is highly specific to Bax and shows no cross-reaction with Bcl-2 or Bcl-X protein. Bcl-2 blocks cell death following a variety of stimuli. Bax has extensive amino acid homology with Bcl-2 and it homodimerizes and forms heterodimers with Bcl-2. CD3, also known as T3, is a member of the immunoglobulin superfamily that plays a role in antigen recognition, signal transduction, and T cell activation. It is found on all mature T-lymphocytes, NK, T-cells, and some thymocytes. It is expressed, typically at high levels, on peripheral T cells and majority of T cell neoplasms. Recognizes a 33 kDa glycoprotein, identified as Nucleophosmin (NPM). It is predominantly localized in the nucleus of cells in most tissues. NPM is involved in ribosomal assembly and rRNA transport. It is an abundant protein that is highly phosphorylated by Cdc2 kinase during mitosis. This phosphoprotein moves between the nucleus and the cytoplasm. It is thought to be involved in several processes including regulation of the ARF/p53 pathway. A number of genes are fusion partners, in particular the anaplastic lymphoma kinase gene on chromosome 2. Mutations in exon 12 affecting the C-terminus of the protein are associated with an aberrant cytoplasmic location. Mutations in this gene are associated with acute myeloid leukemia. Recognizes a protein of 30 kDa which is identified as Prohibitin, an evolutionarily conserved protein with homologues found in yeast to man. It is located in the inner membrane of mitochondria. Although prohibitin mRNA and protein expression occurs throughout the cell cycle, maximum levels are detected during the G1/S phase transition and minimum levels are seen in S phase and the G2/mitosis boundary. Prohibitin is located exclusively in the mitochondria with the highest concentration on the inner membrane. Prohibitin is an ideal mitochondrial marker. The wild-type anaplastic lymphoma kinase (ALK) protein is a 200 kDa transmembrane receptor tyrosine kinase.
