BRCA1 is a nuclear phosphoprotein that plays a role in maintaining genomic stability, and it also acts as a tumor suppressor. It combines with other tumor suppressors, DNA damage sensors, and signal transducers to form a large multi-subunit protein complex known as the BRCA1-associated genome surveillance complex (BASC). BRCA1 associates with RNA polymerase II, and through the C-terminal domain, also interacts with histone deacetylase complexes. This protein thus plays a role in transcription, DNA repair of double-stranded breaks, and recombination. Mutations in this gene are responsible for approximately 40% of inherited breast cancers and more than 80% of inherited breast and ovarian cancers. Thyroperoxidase (TPO) is a 933-amino acid, type I transmembrane glycoprotein that plays a key role in thyroid gland function and autoimmunity. It is present as a dimer on the apical surface of thyroid follicular cells. TPO functions in the iodination of tyrosine residues in thyroglobulin and phenoxy-ester formation between pairs of iodinated tyrosines to generate the thyroid hormones, thyroxine and triiodothyronine. Mutations in this gene are associated with several disorders of thyroid hormonogenesis, including congenital hypothyroidism, congenital goiter, and thyroid hormone organification defect IIA. Malignant thyroid tumors exhibit an anomaly in TPO resulting in lower affinity for anti-TPO. Chromogranin A is present in neuroendocrine cells throughout the body, including the neuroendocrine cells of the large and small intestine, adrenal medulla and pancreatic islets. It is an excellent marker for carcinoid tumors, pheochromocytomas, paragangliomas, and other neuroendocrine tumors. Co-expression of chromogranin A and neuron specific enolase (NSE) is common in neuroendocrine neoplasms. Reportedly, co-expression of certain keratins and chromogranin indicates neuroendocrine lineage. The presence of strong anti-chromogranin staining and absence of anti-keratin staining should raise the possibility of paraganglioma. The co-expression of chromogranin and NSE is typical of neuroendocrine neoplasms. This antibody recognizes an epitope encoded by exon v4 on the variant portion of human CD44. The CD44 molecule belongs to a family of cellular adhesion molecules found on a wide range of normal and malignant cells in epithelial, mesothelial and hemopoiesis tissues. CD44 is a single gene with 20 exons, of which 10 are normally expressed to encode the basic CD44 (H-CAM) molecule. The additional 10 exons (v1 to v10) are only expressed by alternative splicing of the nuclear RNA. This product is prepared by labeling highly cross-adsorbed donkey anti-mouse IgG (H+L) with CF488A dye. CF dyes are Biotium's line of next-generation fluorescent dyes with advantages in brightness, photostability, and conjugate specificity compared to other fluorescent dyes. Green fluorescent CF488A dye has excitation/emission at 490/515 nm. To minimize cross-reactivity, the antibody has been adsorbed against bovine, chicken, goat, guinea pig, Syrian hamster, horse, human, rabbit, and sheep serum.

Donkeys were immunized with highly purified mouse IgG h+l and resulting antiserum was collected. Antibody was solid phase adsorbed using bovine, chicken, goat, guinea pig, horse, hamster, human, rabbit, rat and sheep immunosorbents to remove cross reactive antibody. Specific antibody was immunoaffinity purified off a mouse IgG containing immunosorbent. Antibody concentration was determined using an absorbance at 280 nm: 1.4 equals 1.0 mg of IgG. This antibody will react with mouse IgG and with light chains common with other mouse immunoglobulins as determined by ELISA and IEP techniques. It has less than 1% cross reactivity with bovine, goat, guinea pig, horse, hamster, human, rabbit, rat and sheep IgG and chicken IgY. This antibody is suitable for blotting, ELISA and IHC applications. This antibody may cross react with IgG from other species. Optimal working dilutions should be determined experimentally by the investigator.

To overcome this difficulty the Aurion Series of Secondary ImmunoGold conjugates is based on antibodies raised in Donkey, including Donkey-anti-Goat. It is then known that mixing Donkey-anti-Goat and Donkey-anti-Mouse provides a successful solution.
