Alpha actinins belong to the spectrin gene superfamily which represents a diverse group of cytoskeletal proteins, including the alpha and beta spectrins and dystrophins. Alpha actinin is an actin-binding protein with multiple roles in different cell types. In non-muscle cells, the cytoskeletal isoform is found along microfilament bundles and adherens-type junctions, where it is involved in binding actin to the membrane. In contrast, skeletal, cardiac, and smooth muscle isoforms are localized to the Z-disc and analogous dense bodies, where they help anchor the myofibrillar actin filaments. This gene encodes a muscle-specific, alpha actinin isoform that is expressed in both skeletal and cardiac muscles. This antibody recognizes an intermediate filament protein (IFP) of 55 kDa, which is identified as cytokeratin 7. This MAb is highly specific to cytokeratin 7 and shows no cross-reaction with other IFPs. Cytokeratin 7 is a basic cytokeratin, which is found in most glandular and transitional epithelia but not in the stratified squamous epithelia. Keratin 7 is expressed in the epithelial cells of ovary, lung, and breast but not of colon, prostate, or gastrointestinal tract. This MAb is highly useful in distinguishing ovarian carcinomas (keratin 7 ) from colon carcinomas (keratin 7-).Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. RAD51 is one of the key factors of DNA repair by homologous recombination and has been shown to have anti-apoptotic activity in tumor cells. RAD51 protein interacts with a variety of tumor suppressor proteins including p53, BRCA1 and BRCA2. Elevated expression of RAD51 enhances radio-resistance of human tumor cells. Overexpression of RAD51 protein in tumor cells renders them resistant against cytotoxic drugs like Cisplatin. RAD51 interacts with BRCA1 and BRCA2 to influence subcellular localization and cellular response to DNA damage. BRCA2 inactivation may be a key event leading to genomic instability and tumorigenesis from deregulation of RAD51. High-level expression of RAD51 has been observed in a variety of human malignancies. RAD51 overexpression correlates with histological grading of the tumor in invasive ductal mammary carcinoma. Recognizes a protein of 130 kDa, identified as Desmoglein-3 (DSG3). This MAb is highly specific to Desmoglein-3 and does not cross-react with other members of the Desmoglein-family. DSG3 is a calcium-binding transmembrane glycoprotein component of desmosomes in vertebrate epithelial cells. Research has shown that DSG3 has a very high sensitivity (80%) and specificity (100%) in recognizing squamous cell carcinoma (SqCC). Therefore, DSG3 is considered a very important marker for lung SqCC and can be a useful ancillary marker to separate SqCC from other subtypes of lung cancer. Cytokeratin 13 (KRT13) is the major acidic keratin, which together with KRT4, its basic partner, is expressed in the suprabasal layers of non-cornified stratified epithelia including tongue mucosa, esophagus, anal canal epithelium, tracheal epithelium, uterine cervix, and urothelium. Defects in the KRT13 gene are a cause of white sponge nevus of cannon (WSN), a rare autosomal dominant disorder, which predominantly affects non-cornified stratified squamous epithelia and is characterized by the presence of soft, white and spongy plaques in the oral mucosa. KRT13 has been used as a marker for non-keratinized squamous epithelium. It is also expressed in various squamous metaplasia, but it is down regulated in squamous dysplasia and squamous carcinoma. This antibody recognizes a protein of 40 kDa, identified as Thyroid transcription factor-1 (TTF-1). TTF-1 is a member of the NKx2 family of homeodomain transcription factors. It is expressed in epithelial cells of the thyroid gland and the lung. Nuclei from liver, stomach, pancreas, small intestine, colon, kidney, breast, skin, testes, pituitary, prostate, and adrenal glands are unreactive. Anti-TTF-1 is useful in differentiating primary adenocarcinoma of the lung from metastatic carcinomas originating in the breast, mediastinal germ cell tumors, and malignant mesothelioma. It can also be used to differentiate small cell lung carcinoma from lymphoid infiltrates. Loss of TTF-1 expression in non-small cell lung carcinoma has been associated with aggressive behavior of such neoplasms. This antibody recognizes a protein of ~150 kDa, identified as Desmoglein-1 (DSG1). Desmoglein-1 is a member of the desmosomal cadherin family. Desmosomes are intercellular adhering junctions that represent cell surface attachment sites for intermediate filament. Desmocollins and desmogleins are the main desmosomal transmembrane proteins. Desmogleins consist of Dsg1, Dsg2, Dsg3, and Dsg4 isoforms. Within the desmosome, the extracellular domain of desmoglein is essential for calcium dependent heterophilic binding to the desmocollins, whereas the intracellular domain is essential for binding to the desmosomal plaque protein, plakoglobin. Desmoglein 1 is synthesized exclusively in the suprabasal layers.
