APEX Nuclease (also known as APEX1 or APE1) is a multifunctional protein that plays a central role in the cellular response to oxidative stress. The two major activities of APEX1 are in DNA repair and redox regulation of transcriptional factors. It functions as a apurinic/apyrimidinic (AP) endodeoxyribonuclease in the DNA base excision repair (BER) pathway of DNA lesions induced by oxidative and alkylating agents. Patients with genetic variants in APEX1 have been shown to have a higher risk of lung cancer. Elevated APEX1 levels observed in human testicular cancer may be related to relative resistance to therapy and therefore may serve as a diagnostic marker for refractory disease. Recognizes a protein of 40 kDa, identified as CD7 (also known as gp40, Leu9). CD7 is a member of the immunoglobulin gene superfamily. Its N-terminal amino acids 1-107 are highly homologous to Ig kappa-L chains whereas the carboxyl-terminal region of the extracellular domain is proline-rich and has been postulated to form a stalk from which the Ig domain projects. CD7 is expressed on the majority of immature and mature T-lymphocytes, and T cell leukemia. It is also found on natural killer cells, a small subpopulation of normal B cells and on malignant B cells. Cross-linking surface CD7 positively modulates T cell and NK cell activity as measured by calcium fluxes, expression of adhesion molecules, cytokine secretion and proliferation. CD7 associates directly with phosphoinositol 3'-kinase. CD7 ligation induces production of D-3 phosphoinositides and tyrosine phosphorylation. Recognizes a protein of 54 kDa, which is identified as cyclin A. Its epitope is located amino acids 144-148 of human Cyclin A2. Cyclins are regulatory subunits of the cyclin-dependent kinases (cdk's) and they control transition at different specific phases of the cell cycle. The temporal expression of cyclins is tightly regulated and subsequently plays a critical role in controlling the enzymatic activity of cdk's. These cyclin/cdk complexes are essential for passage through specific stages in the cell cycle. In mammalian somatic cells, cyclin A is required for S-phase and passage through G2-phase. The D and E type cyclins regulate the passage of G1, while cyclin B is a critical regulator of mitosis. Mutation or disruption of normal cyclin A expression causes cells to arrest in G2-phase. PD-1 is expressed on a subset of CD4-CD8-thymocytes, and on activated T and B cells. PD-1 is thought to be involved in lymphocyte clonal selection and peripheral tolerance. The PD-1 ligands, PD-L1 (also known as B7-H1) and PD-L2 (B7-DC), are members of the B7 immunoglobulin superfamily. PD-1 is transiently expressed on CD4 and CD8 thymocytes as well as activated T and B-lymphocytes and myeloid cells. Additionally, Pdcd1mRNA is expressed in developing B-lymphocytes during the pro-B-cell stage. Podocalyxin is a member of the CD34 transmembrane sialomucin family. It is over-expressed on the podocyte foot projections and plays essential roles in kidney development and homeostasis, blood filtration and urine formation. It is also expressed on vascular endothelia, hematopoietic progenitors and a subset of neurons. Overexpression of podocalyxin may be linked to more aggressive tumor behavior. Podocalyxin antibody can identify podocytes in the urine (podocyturia) that may indicate glomerular disease, pre-eclampsia, and other kidney pathology. This antibody recognizes the Calcineurin B alpha protein, encoded by the PPP3R1 (Protein Phosphatase 3 Regulatory Subunit B Alpha) gene. Calcineurin is an enzyme that dephosphorylates serine and threonine residues in proteins. It is a heterodimer of a 59 kDa catalytic A subunit and a 19 kDa regulatory B subunit that is activated by the binding of calcium ions and calmodulin. Calcineurin is expressed in many tissues, but its levels are highest in the brain, where it may play a role in learning and memory. Recognizes a protein of 110 kDa, identified as Excision Repair Cross Complementing 1 (ERCC1). It is a mammalian nucleotide excision repair (NER) enzyme involved in repair of damaged DNA. ERCC1 is a homologous to RAD10 in Saccharomyces cerevisiae, which is required in mitotic intrachromosomal recombination and repair. ERCC1 is required in repair of cisplatin-induced DNA adducts and ultraviolet (UV)-induced DNA damage. Calbindin 1, also known as calbindin D28K, is a member of a family of calcium binding proteins. This protein family also includes calmodulin, calbindin 2, S-100, and troponin C. Calbindin 1 is a 261-amino acid protein with 6 EF-hand domains, 4 of which are active calcium-binding domains. Expressed in brain, ovary, uterus, testis, pancreas, liver, kidney and intestine, calbindin 1 acts as a calcium-buffering agent and alters the activity of the plasma membrane ATPase. In neuronal cells, calbindin 1 modulates calcium channel activity, calcium transients and intrinsic neuronal firing activity. This gene encodes an enzyme that transfers methyl groups to cytosine nucleotides of genomic DNA. This protein is the major enzyme responsible for maintaining methylation patterns following DNA replication and shows a preference for hemi-methylated DNA. Recognizes an epitope located on collagenase digested Type VII collagen (150 kDa), i. e. the non-helical carboxyl terminal region of the dimer. EBAG9, also known as RCAS1, is an estrogen-transcribed protein. Soluble and membranous RCAS1 proteins may play a role in the immune escape of tumor cells by promoting T lymphocyte inhibition of growth and apoptosis. RCAS1 is expressed in a wide variety of cancers, including uterine, ovarian, and lung cancer cells, and acts as a ligand for a putative receptor present on peripheral lymphocytes. RCAS1 is highly expressed not only in cancer cells but also in non-tumor bile duct cells subject to immune attack.
