Cytokeratins are a family of intermediate filament proteins that assemble into filaments through forming heterodimers of one type I Cytokeratins (Cytokeratins 9 to 23) and one type II Cytokeratins (keratins 1 to 8). The cytokeratin proteins play a critical role in differentiation, as well as tissue specialization and function, to maintain the overall structural integrity of epithelial cells. Cytokeratins are also useful markers in identifying the origin of metastatic tumors. Cytokeratin 16 is expressed in benign stratified squamous epithelium and squamous cell carcinoma of the head and neck, as well as luminal cells of mammary gland and sweat ducts. This antibody recognizes an epitope encoded by exon v4 on the variant portion of human CD44. The CD44 molecule belongs to a family of cellular adhesion molecules found on a wide range of normal and malignant cells in epithelial, mesothelial and hemopoiesis tissues. CD44 is a single gene with 20 exons, of which 10 are normally expressed to encode the basic CD44 (H-CAM) molecule. The additional 10 exons (v1 to v10) are only expressed by alternative splicing of the nuclear RNA. The expression of specific cell adhesion molecule CD44 splice variants has been reported to be associated with metastasis in certain human malignancies. This MAb recognizes a protein of 56.5 kDa, identified as cytokeratin 10 (CK10). CK10 is expressed in all suprabasal layers of the epidermis. In the epidermis, expression of CK10 strictly parallels the extent of differentiation; it is absent in the basal layer, appears in the first suprabasal layers and increases in concentration towards the granular layer. However, CK10 is rarely detected in early stages of vulvar squamous carcinomas (tumors less than 2 cm, clinical stage I) regardless of the tumor grade. In larger and more advanced tumors (greater than 2 cm, clinical stages II and III), CK10 is detected very frequently. This MAb is specific to lambda light chain of immunoglobulin and shows no cross-reaction with lambda light chain or any of the five heavy chains. In mammals, the two light chains in an antibody are always identical, with only one type of light chain, kappa or lambda. The ratio of Kappa to Lambda is 70:30. However, with the occurrence of multiple myeloma or other B-cell malignancies this ratio is disturbed. Antibody to the lambda light chain is reportedly useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. B-cell lymphoma/leukemia 10 (BCL10) is a signaling protein that promotes apoptosis, pro-caspase-9 maturation, and activation of NF-kappa-B via NIK and IKK. BCL10 contains an N-terminal caspase recruitment domain (CARD), which is found in a number of apoptotic regulatory molecules. BCL10 is involved in the adaptive immune response, and may be an adapter protein between upstream TNFR1-TRADD-RIP complex and the downstream NIK-IKK-IKAP complex. A BCL10 translocation is recurrent in low-grade mucosa-associated lymphoid tissue (MALT lymphoma). The matrix metalloproteinases (MMP) are a family of peptidase enzymes responsible for the degradation of extracellular matrix components, including collagen, gelatin, fibronectin, laminin and proteoglycan. PMEPA1 (prostate transmembrane protein, androgen induced 1 is a 287 amino acid single-pass membrane protein that contains WW-binding motifs and localizes to the cell membrane. Expressed at high levels in prostate, kidney and ovary, PMEPA1 interacts with NEDD4 and may play a role in regulating AR (androgen receptor) levels, specifically in prostate cells.
