Activation of TAL1 characterizes up to 60% of cases of human T cell acute lymphoblastic leukemia, making it the most frequent gain-of-function mutation observed in this disorder. TAL1 (also designated SCL) is a serine phosphoprotein and basic helix-loop-helix transcription factor known to regulate embryonic hematopoiesis. This transcription factor binds as a heterodimer with E2A and HEB/HTF4 to a nucleotide sequence motif termed the E-box. In addition, leukemogenesis is accelerated dramatically by transgenic co-expression of TAL1 and the catalytic subunit of casein kinase IIHLH transcription factors. Drebrins (developmentally regulated brain proteins) are cytoplasmic proteins that bind F-actin in the brain and are involved in cell migration, extension of neuronal processes and plasticity of dendrites. There are three isoforms: two embryonic types (E1 and E2); and an adult type (A), generated by alternative RNA splicing from a single Drebrin gene. Drebrins are expressed mainly in brain neurons but are also found in skeletal muscle, heart, placenta, pancreas and kidney. Drebrin has been designated as a marker of the dendritic spine. This antibody reacts with Ig domain of CD47 protein. CD47, originally named integrin-associated protein (IAP), is a 50 kDa protein containing five membrane-spanning sequences and a short cytoplasmic tail. CD47 plays a role in both cell adhesion by acting as an adhesion receptor for THBS1 on platelets, and in the modulation of integrins. It is important in memory formation and synaptic plasticity in the hippocampus. CD47 may play a role in membrane transport and/or integrin dependent signal transduction. Cathepsin K is a lysosomal and secreted cysteine proteinase involved in bone remodeling and resorption. This protein, which is a member of the peptidase C1 protein family, is predominantly expressed in osteoclasts. However, the encoded protein is also expressed in a significant fraction of human breast cancers, where it could contribute to tumor invasiveness. Mutations in this gene are the cause of pycnodysostosis, an autosomal recessive disease characterized by osteosclerosis and short stature. Expression of DOG-1 protein is elevated in the gastrointestinal stromal tumors (GIST's), c-kit signaling-driven mesenchymal tumors of the GI tract. DOG-1 is rarely expressed in other soft tissue tumors, which, due to appearance, may be difficult to diagnose. Immunoreactivity for DOG-1 has been reported in 97. 8 percent of scorable GIST's, including all c-kit negative GIST's. Recognizes a protein of 130 kDa, identified as Desmoglein-3 (DSG3). This MAb is highly specific to Desmoglein-3 and does not cross-react with other members of the Desmoglein-family. DSG3 is a calcium-binding transmembrane glycoprotein component of desmosomes in vertebrate epithelial cells. Research has shown that DSG3 has a very high sensitivity (80%) and specificity (100%) in recognizing squamous cell carcinoma (SqCC). Therefore, DSG3 is considered a very important marker for lung SqCC and can be a useful ancillary marker to separate SqCC from other subtypes of lung cancer. Recognizes a 33 kDa glycoprotein, identified as Nucleophosmin (NPM). It is predominantly localized in the nucleus of cells in most tissues. NPM is involved in ribosomal assembly and rRNA transport. It is an abundant protein that is highly phosphorylated by Cdc2 kinase during mitosis. This phosphoprotein moves between the nucleus and the cytoplasm. It is thought to be involved in several processes including regulation of the ARF/p53 pathway. A number of genes are fusion partners, in particular the anaplastic lymphoma kinase gene on chromosome 2. Mutations in exon 12 affecting the C-terminus of the protein are associated with an aberrant cytoplasmic location. Mutations in this gene are associated with acute myeloid leukemia. The initiation of DNA replication is a multi-step process that depends on the formation of pre-replication complexes, which trigger initiation. Among the proteins required for establishing these complexes are the origin recognition complex (ORC) proteins. ORC proteins bind specifically to origins of replication where they serve as scaffold for the assembly of additional initiation factors. Human ORC subunits 1-6 are expressed in the nucleus of proliferating cells and tissues, such as the testis. Myofibroblasts are a unique group of smooth muscle-like fibroblasts that play an important role in oncogenesis, inflammation, repair, wound contraction and fibrosis. Like smooth muscle (SM) cells, myofibroblasts contain microfilament bundles and express -SM Actin, the Actin isoform that is present in myoepithelial cells and SM cells and especially abundant in vascular SM cells. Myofibroblasts secrete inflammatory and anti-inflammatory cytokines, chemokines, growth factors and lipid and gaseous inflammatory mediators, as well as extracellular matrix proteins and proteases in most organs and tissues.
