DNA methylation is the major modification of eukaryotic genomes and plays an essential role in mammalian development. Human proteins MECP2, MBD1, MBD2, MBD3, and MBD4 comprise a family of nuclear proteins related by the presence in each of a methyl-CpG binding domain (MBD). Each of these proteins, with the exception of MBD3, is capable of binding specifically to methylated DNA. MECP2, MBD1 and MBD2 can also repress transcription from methylated gene promoters. MBD1 has also been predicted to be involved in the development and progression of tumors, and it has been shown that MBD1 is significantly upregulated in pancreatic cancer tissues when compared with surrounding normal tissues. EBAG9, also known as RCAS1, is an estrogen-transcribed protein. Soluble and membranous RCAS1 proteins may play a role in the immune escape of tumor cells by promoting T lymphocyte inhibition of growth and apoptosis. RCAS1 is expressed in a wide variety of cancers, including uterine, ovarian, and lung cancer cells, and acts as a ligand for a putative receptor present on peripheral lymphocytes. RCAS1 is highly expressed not only in cancer cells but also in non-tumor bile duct cells subject to immune attack. RCAS1 inhibits the in vitro growth of receptor-expressing cells and induces apoptosis, contributing to the ability of tumor cells to evade host immune surveillance. High expression of RCAS1 significantly correlates with tumor progression and with poor outcome for many cancer patients. This antibody recognizes a protein doublet of 20-22 kDa, identified as MART-1 (Melanoma Antigen Recognized by T cells 1) or Melan-A. MART-1 is a newly identified melanocyte differentiation antigen recognized by autologous cytotoxic T lymphocytes. Seven other melanoma associated antigens recognized by autologous cytotoxic T cells include MAGE-1, MAGE-3, tyrosinase, gp100, gp75, BAGE-1, and GAGE-1. Subcellular fractionation shows that MART-1 is present in melanosomes and endoplasmic reticulum. This MAb labels melanomas and other tumors showing melanocytic differentiation. It is also a useful positive-marker for angiomyolipomas. SOX2 is required for stem cell maintenance in the central nervous system, and it also regulates gene expression in the stomach. SOX2 is necessary for regulating multiple transcription factors that affect Oct 3/4 expression. An essential function of SOX2 is to stabilize embryonic stem cells in a pluripotent state by maintaining the requisite level of Oct 3/4 expression. Reportedly, SOX2 is associated with aggressive phenotypes of breast, head and neck, gastric, colorectal, bladder, and small cell lung cancers. However, SOX2 is expressed in a high percentage of lung squamous cell carcinomas and has been shown to be an independent favorable prognostic marker. Recognizes a protein of 31 kDa, which is identified as MTAP (5'-deoxy-5'-methylthioadenosine phosphorylase). It catalyzes the reversible phosphorolysis of methylthioadenosine, which is important in polyamine metabolism and for the salvage of adenine and methionine. The gene encoding MTAP is linked to the tumor suppressor gene, p16INK4A. Deficient levels of MTAP can occur in cancers primarily through co-deletion of the MTAP gene and the p16INK4A gene. Cells expressing MTAP and possessing adenine salvage pathway activity may be less susceptible to malignancy due to growth-inhibitory actions of agents (e. g. antifolates), whose mechanism of action, in part, involves this de novo purine pathway. Expression of NKX2.2 has been found in neuroendocrine tumors of the gut, making it a potential marker for the study of gastrointestinal neuroendocrine tumors. More recently, NKX2.2 protein was identified as a target of EWS-FLI-1, the fusion protein specific to Ewing sarcoma, and was shown to be differentially upregulated in Ewing sarcoma on the basis of array-based gene expression analysis. HHV 8 encodes a latent nuclear antigen (LNA), which is the product of the viral gene orf 73. LNA is capable of forming a complex with retinoblastoma susceptibility gene product, which may be related to its oncogenic activity. HHV8 is associated with three different diseases observed in AIDS patients; kaposi's sarcoma, primary effusion lymphoma (which is a rare type of non-Hodgkin lymphoma affecting the body cavities) and multicentric Castleman's disease. This MAb recognizes the peptide core of gastric mucin M1 (recently identified as Mucin 5AC). Its epitope is located in the C-terminal cysteine rich part of the peptide core of MUC5AC. Its epitope is destroyed by beta-mercaptoethanol but not by periodate treatment. This mucin is present in primary ovarian mucinous cancer but usually absent in colorectal adenocarcinoma, thus showing an expression pattern opposite to MUC2. Together with a panel of antibodies, Anti-MUC5AC may be useful for differential identification of primary mucinous ovarian tumors from colon adenocarcinoma metastatic to the ovary. MUC5AC antibodies may also be useful for identification of intestinal metaplasia as well as in the identification of pancreatic carcinoma and pre-cancerous changes vs. normal pancreas.
