S100 belongs to the family of calcium binding proteins. S100A and S100B proteins are two members of the S100 family. S100A is composed of an alpha and a beta chain whereas S100B is composed of two beta chains. This antibody is specific against an epitope located on the beta-chain (i.e. in S-100A and S-100B) but not on the alpha-chain of S-100 (i.e. in S-100A and S100A0). This antibody can be used to localize S-100A and S-100B in various tissue sections. S-100 protein has been found in normal melanocytes, Langerhans cells, histiocytes, chondrocytes, lipocytes, skeletal and cardiac muscle, Schwann cells, epithelial and myoepithelial cells of the breast, salivary and sweat glands, as well as in glial cells. VISTA / GI24 is a transmembrane protein expressed in bone, on embryonic stem cells (ESCs), and on tumor cell surfaces. On ESC s, Gi24 appears to positively interact with BMP-4, potentiating BMP signaling and the transition from an undifferentiated to a differentiated state. On tumor cells, Gi24 both promotes MT1-MMP expression and activity and serves as a substrate for MT1-MMP. This increases the potential for cell motility. Mature human Gi24 contains a 162aa extracellular region with one V-type Ig-like domain and a 96aa cytoplasmic domain. This antibody recognizes a protein of 104 kDa-110 kDa, characterized as major vault protein (MVP). Vaults are large ribonucleoprotein particles (RNPs) present in all eukaryotic cells. They have a complex morphology, including several small molecules of RNA, but a single protein species. The MVP accounts for >70% of their mass. Their shape is reminiscent of the nucleopore central plug. Treatment of cells with estradiol increases the amount of MVP in nuclear extract. The hormone-dependent interaction of vaults with ER is prevented in vitro by sodium molybdate. Antibodies to estrogen, progesterone and glucocorticoid receptors are able to co-immunoprecipitate the MVP. MVP is overexpressed in many neoplastic tissues and cell lines. GP2 (glycoprotein 2), also known as ZAP75, is a 537 amino acid secreted protein. It is an integral membrane protein that is secreted from intracellular zymogen granules and associates with the plasma membrane via glycosylphosphatidylinositol (GPI) linkage. GP2 is cleaved and then released into the pancreatic duct along with exocrine secretions. GP2 binds pathogens such as enterobacteria, thereby playing an important role in the innate immune response. The C-terminus of this protein is related to the C-terminus of the protein encoded by the neighboring gene, uromodulin (UMOD). GP2 is also expressed on the apical plasma membrane of specialized microfold (M) cells among enterocytes and serves as a transcytotic receptor for mucosal antigens. Recognizes a 33 kDa glycoprotein, identified as Nucleophosmin (NPM). It is predominantly localized in the nucleus of cells in most tissues. NPM is involved in ribosomal assembly and rRNA transport. It is an abundant protein that is highly phosphorylated by Cdc2 kinase during mitosis. This phosphoprotein moves between the nucleus and the cytoplasm. It is thought to be involved in several processes including regulation of the ARF/p53 pathway. A number of genes are fusion partners, in particular the anaplastic lymphoma kinase gene on chromosome 2. Mutations in exon 12 affecting the C-terminus of the protein are associated with an aberrant cytoplasmic location. Mutations in this gene are associated with acute myeloid leukemia. This antibody recognizes a protein of 145 kDa, identified as CD11c. CD11c (ITGAX), a member of the leukointegrin family, shares the same beta subunit with other members of the leukocyte adhesion molecule family, which includes CD11a (LFA-1), CD11b (MAC-1) and CD11d (ITGAD), but has a unique alpha chain. CD11c has been shown to play a role in phagocytosis, cell migration, and cytokine production by monocytes/macrophages as well as induction of T-cell proliferation by Langerhans cells. CD11c is expressed prominently on the plasma membranes of monocytes, tissue macrophages, NK cells, and most dendritic cells (DCs). A lower level of expression is also observed on neutrophils as a result of its high level of expression on most DCs. An antibody to CD11c may aid in identification of lesions with histiocytic origin. The intestine-specific transcription factors CDX1 and CDX2 are important for directing intestinal development, differentiation, proliferation and maintenance of the intestinal phenotype. CDX2 protein expression has been seen in GI carcinomas. Anti-CDX2 has been useful to establish GI origin of metastatic adenocarcinomas and carcinoids and is especially useful to distinguish metastatic colorectal adenocarcinoma from lung adenocarcinoma. This MAb reacts with a 68 kDa protein, identified as light sub-unit of neurofilaments (NF-L). Neurofilaments make up the main structural elements of axons and dendrites and are found in neurons, peripheral nerves, and sympathetic ganglion cells. Neurofilaments consist of three major subunits with molecular weights of 68 kDa (NF-L), 160 kDa (NF-M) and 200 kDa (NF-H). von Willebrand Factor (vWF) is a multimeric glycoprotein that is found in endothelial cells, plasma and platelets. It acts as a carrier protein for Factor VIII and promotes platelet adhesion and aggregation. vWF undergoes a variety of posttranslational modifications that influence the affinity and availability for Factor VIII, including cleavage of the propeptide and formation of N-terminal disulfide bonds. This antibody recognizes an epitope encoded by exon v3 on the variant portion of human CD44. The CD44 molecule belongs to a family of cellular adhesion molecules found on a wide range of normal and malignant cells in epithelial, mesothelial and hemopoiesis tissues. CD44 is a single gene with 20 exons, of which 10 are normally expressed to encode the basic CD44 (H-CAM) molecule. The additional 10 exons (v1 to v10) are only expressed by alternative splicing of the nuclear RNA.
