The Lewis histo-blood group system comprises a set of fucosylated glycosphingolipids that are synthesized by exocrine epithelial cells and circulate in body fluids. The glycosphingolipids function in embryogenesis, tissue differentiation, tumor metastasis, inflammation, and bacterial adhesion. They are secondarily absorbed to red blood cells giving rise to their Lewis phenotype. This gene is a member of the fucosyltransferase family, which catalyzes the addition of fucose to precursor polysaccharides in the last step of Lewis antigen biosynthesis. It encodes an enzyme with alpha(1,3)-fucosyltransferase and alpha(1,4)-fucosyltransferase activities. Lewis blood group antigens are carbohydrate moieties structurally integrated in mucous secretions. Lewis antigen system alterations have been described in gastric carcinoma and associated lesions. Recognizes a protein of 66 kDa-170 kDa, identified as involucrin. In Western blotting of cultured human keratinocytes, this MAb reacts with a 120 kDa protein. It stains the involucrin in a variety of sizes: 170 kDa in MCF-7 cells, a doublet of ~115 kDa and 150 kDa in gorilla and owl monkey, 66 kDa in dog, and a doublet of 105 kDa in pig. Its epitope maps between codon 421-568 of human involucrin. Involucrin is expressed in a range of stratified squamous epithelia, including the cornea, which lacks a distinct cornified layer. In normal epidermis, it is first expressed in the upper spinous layers, and in keratinocyte cultures, all cells that have left the basal layer express it. The major constituents of mucus, the viscous secretion that covers epithelial surfaces such as those in the trachea, colon, and cervix, are highly glycosylated proteins called mucins. These glycoproteins play important roles in the protection of the epithelial cells and have been implicated in epithelial renewal and differentiation. This gene encodes an integral membrane glycoprotein found on the cell surface, although secreted isoforms may exist. MUC-4 transcripts have been detected in normal respiratory epithelium and lung. MUC-4 is a very specific (100%) and sensitive (90%) marker of lung adenocarcinomas and is negative for mesotheliomas. Reportedly, MUC-4 expression in invasive ductal carcinoma of the pancreas is an independent factor for poor prognosis. Accumulating evidence indicates that NuRD may regulate the transcription of specific genes by interacting with specific transcriptional factors. In addition, the NuRD complex may also participate in genome-wide transcriptional regulation through an association with histone tails. Recognizes a protein of 55-62 kDa, identified as cyclin B1. In mammals, cyclin B associates with inactive p34cdc2, which facilitates phosphorylation of p34cdc2 at aa 14Thr and 15Tyr. This maintains the inactive state until the end of G2-phase. The inactive cyclin B-p34cdc2 complex continues to accumulate in the cytoplasm until the completion of DNA synthesis, when Cdc25, a specific protein phosphatase, dephosphorylates aa 14Thr and 15Tyr of p34cdc2 rendering the complex active at the G2/M boundary. This mitotic kinase complex remains active until the metaphase/anaphase transition when cyclin B is degraded. This degradation process is ubiquitin-dependent and is necessary for the cell to exit mitosis. So, cyclin B-p34cdc2 plays a critical role in G2 to M transition. Keratin 15 is a type I keratin which is expressed only in basal keratinocytes in stratified epithelia and does not appear to have a natural type II expression partner. Keratin 15 is down regulated in activated keratinocytes. Cytokeratin 15 is a specific marker of stem cells of the hair-follicle bulge and may be a useful marker for diagnosis between basal cell carcinoma (BCC) and trichoepithelioma. Trichoblastoma are benign neoplasms of follicular differentiation frequently found in nevus sebaceous. Many morphologic features are shared with nodular basal cell carcinoma, sometimes rendering a diagnosis difficult. This antibody recognizes a glycoprotein of 75 kDa, identified as low affinity Nerve Growth Factor (NGF) Receptor (p75NGFR) or Neurotrophin Receptor (p75NTR). NGFR is expressed in various neural crest cells and their tumors such as melanocytes, melanomas, neuroblastomas, pheochromocytomas and neurofibromas. Reportedly, anti-NGFR is a reliable marker for desmoplastic and neurotropic melanomas. Myoglobin is a cytosolic oxygen-binding protein responsible for the storage and diffusion of oxygen within myocytes. Expression of myoglobin is highest in skeletal and cardiac muscle. Napsin is a pepsin-like aspartic proteinase connected with maturation of surfactant protein B. There are two closely related napsins, napsin A and napsin B. Napsin A is expressed as a single chain protein. Immunohistochemical studies revealed high expression levels of napsin A in human lung and kidney but low expression in spleen. Napsin A is expressed in type II pneumocytes and in adenocarcinomas of lung. This antibody recognizes a 32 kDa protein, which is identified as homodimer of galectin-13 (also known as PP13). Galectins are a family of soluble beta-galactoside-binding lectins that modulate cell-to-cell adhesion and cell-to-extracellular matrix (ECM) interactions and play a role in tumor progression, pre-mRNA splicing and apoptosis. One such member, galectin-13, is a 139 amino acid protein that contains one galectin domain. Possessing lysophospholipase activity, galectin-13 exists as a disulfide-linked homodimer.
