Chromogranin A is present in neuroendocrine cells throughout the body, including the neuroendocrine cells of the large and small intestine, adrenal medulla and pancreatic islets. It is an excellent marker for carcinoid tumors, pheochromocytomas, paragangliomas, and other neuroendocrine tumors. Co-expression of chromogranin A and neuron specific enolase (NSE) is common in neuroendocrine neoplasms. Reportedly, co-expression of certain keratins and chromogranin indicates neuroendocrine lineage. The presence of strong anti-chromogranin staining and absence of anti-keratin staining should raise the possibility of paraganglioma. The co-expression of chromogranin and NSE is typical of neuroendocrine neoplasms. Biotium Major Vault Protein (MVP)(1032), CF740 conjugate, 0.1mg/mL
Recognizes a protein of 104 kDa-110 kDa, characterized as major vault protein (MVP). Vaults are large ribonucleoprotein particles (RNPs) present in all eukaryotic cells. They have a complex morphology, including several small molecules of RNA, but a single protein species. The MVP accounts for >70% of their mass. Their shape is reminiscent of the nucleopore central plug. Treatment of cells with estradiol increases the amount of MVP in nuclear extract. The hormone-dependent interaction of vaults with ER is prevented in vitro by sodium molybdate. Antibodies to estrogen, progesterone and glucocorticoid receptors are able to co-immunoprecipitate the MVP. MVP is overexpressed in many neoplastic tissues and cell lines. This antibody recognizes a carbohydrate epitope present on tumor-associated Lewis Y antigen (Fucalpha1-2Galbeta1-4/3GlcNAcbeta). Lewis Y is expressed in large bowel tumors and colorectal carcinomas. It may be useful in the classification of human renal and bladder tumors. The Lewis Y antigen has been evaluated as a clinical marker for the diagnosis and prognosis of cholangiocarcinoma, hepatocellular carcinoma and breast cancer.Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. Calpain 1 is an intracellular calcium-dependent protease that cleaves cytoskeletal and sub-membranous proteins. Calpains are non-lysosomal, calcium-activated intracellular cysteine proteases. Calpains mediate specific Ca2 - dependent processes including cell fusion, mitosis and meiosis. Calpains are heterodimers of a small regulatory subunit and one of three large catalytic subunits, designated Calpain 1, Calpain 2 and Calpain p94. Calpastatin regulates Calpain by inhibiting both the proteolytic activity of Calpain and its binding to membranes. Calpastatin exists in two types, tissue type and erythrocyte type, resulting from both alternative splicing and proteolytic processing. Calpain 1 co-localizes with human leukocyte antigen-DR (HLA-DR) on activated microglia in the aging brain. Perilipin-2, also known as Adipophilin, belongs to the perilipin family, members of which coat intracellular lipid storage droplets. This 48 kDa protein is associated with the lipid globule surface membrane material, and maybe involved in development and maintenance of adipose tissue. This MAb reacts with MUC1, a large transmembrane glycoprotein expressed on the ductal surface of normal glandular epithelia. It is used as tracer agent in CA15. 3 assays. The extracellular domain of MUC1 largely consists of a highly conserved, O-glycosylated 20 amino acids tandem repeat which can occur 30-100 times per molecule depending on the length of the allele involved. In the vast majority of human carcinomas this protein is up-regulated and poorly glycosylated and appears on the cell surface in a non-polarized fashion. The dominant epitope of this MAb involves both amino acids as well as sugar moieties. Its epitope is destroyed by desialylation i. e. treatment with Neuraminidase.
