Recognizes a protein of 75 kDa, identified as mu heavy chain of human immunoglobulins. This antibody reacts with human leukemia cell line HL-60 and immunoprecipitates two proteins with MW of 48 kDa and 52 kDa. It does not stain B-cell lines Daudi, PHS, Namalwa, RPMI-1788 and T-cell lines CCRF-HSB2, Jurkat and Molt-4. This antibody stains monocytes and up to 10% of lymphocytes from peripheral blood of healthy donors. Blast cells of patients with AMMonL (M5 following FAB classification), AMMonL (M4) and hairy cells leukemia are positive. Its antigen is particularly expressed on blood cells from patients with infectious mononucleosis and CLL. Histiocytes and macrophages are also positive. This antibody recognizes a protein of 30-33 kDa, which is identified as CD20. Its epitope is located in the cytoplasmic domain of CD20 and was, therefore, ascribed as CD20cy in the 5th Workshop. CD20 is a non-Ig differentiation antigen of B-cells and its expression is restricted to normal and neoplastic B-cells, being absent from all other leukocytes and tissues. CD20 is expressed by pre B-cells and persists during all stages of B-cell maturation but is lost upon terminal differentiation into plasma cells.
