GATA-3 is a zinc finger transcription factor and plays an important role in promoting and directing cell proliferation, development, and differentiation in many tissues and cell types. GATA-3 expression is primarily seen in breast and urothelial carcinomas. 'Therefore, GATA3 antibody can be useful in the identification of unknown primary carcinoma when carcinomas of the breast or bladder are a possibility. RAD51 is one of the key factors of DNA repair by homologous recombination and has been shown to have anti-apoptotic activity in tumor cells. RAD51 protein interacts with a variety of tumor suppressor proteins including p53, BRCA1 and BRCA2. Elevated expression of RAD51 enhances radio-resistance of human tumor cells. Overexpression of RAD51 protein in tumor cells renders them resistant against cytotoxic drugs like Cisplatin. RAD51 interacts with BRCA1 and BRCA2 to influence subcellular localization and cellular response to DNA damage. BRCA2 inactivation may be a key event leading to genomic instability and tumorigenesis from deregulation of RAD51. High-level expression of RAD51 has been observed in a variety of human malignancies. This antibody recognizes a protein of 26 kDa, identified as CD81 (Workshop VI; Code CD81.1). CD81 has a very broad cellular distribution, being expressed on T- and B-lymphocytes, NK cells, thymocytes, eosinophils, fibroblasts, epithelial and endothelial cells. Neutrophils, erythrocytes and platelets are negative, while monocytes are variably positive. CD81 is a member of a family of tetraspanin transmembrane proteins, including CD9, CD37, CD53, CD63, and CD82. It associates with CD19, CD21, Leu 13, and integrins on cell membrane and is involved in signal transduction in B lymphocyte development and cell adhesion. CD81 also acts as a receptor for the envelope protein E2 of chronic hepatitis C virus. This MAb is specific to progesterone receptor and shows minimal cross-reaction with other members of the family. Progesterone receptor is expressed as two major isoforms, PR-A (81 kDa) and PR-B (116 kDa). This antibody recognizes a protein of 40 kDa, identified as Thyroid transcription factor-1 (TTF-1). TTF-1 is a member of the NKx2 family of homeodomain transcription factors. It is expressed in epithelial cells of the thyroid gland and the lung. Nuclei from liver, stomach, pancreas, small intestine, colon, kidney, breast, skin, testes, pituitary, prostate, and adrenal glands are unreactive. Anti-TTF-1 is useful in differentiating primary adenocarcinoma of the lung from metastatic carcinomas originating in the breast, mediastinal germ cell tumors, and malignant mesothelioma. It can also be used to differentiate small cell lung carcinoma from lymphoid infiltrates. Loss of TTF-1 expression in non-small cell lung carcinoma has been associated with aggressive behavior of such neoplasms. This MAb reacts with a 200 kDa and 68 kDa protein, identified as heavy and light sub-units of neurofilaments (NF-H NF-L). Neurofilaments make up the main structural elements of axons and dendrites and are found in neurons, peripheral nerves, and sympathetic ganglion cells. Neurofilaments consist of three major subunits with molecular weights of 68 kDa (NF-L), 160 kDa (NF-M) and 200 kDa (NF-H). Anti-neurofilament stains a number of neural, neuroendocrine, and endocrine tumors. Neuromas, ganglioneuromas, gangliogliomas, ganglioneuroblastomas, and neuroblastomas stain positively for anti-neurofilament. Neurofilaments are also present in paragangliomas as well as adrenal and extra-adrenal pheochromocytomas. GP2 (glycoprotein 2), also known as ZAP75, is a 537 amino acid secreted protein. It is an integral membrane protein that is secreted from intracellular zymogen granules and associates with the plasma membrane via glycosylphosphatidylinositol (GPI) linkage. GP2 is cleaved and then released into the pancreatic duct along with exocrine secretions. GP2 binds pathogens such as enterobacteria, thereby playing an important role in the innate immune response. The C-terminus of this protein is related to the C-terminus of the protein encoded by the neighboring gene, uromodulin (UMOD). This antibody is a mixture of four different monoclonal antibodies. This antibody is specific to 60 kDa latent membrane protein (LMP-1) encoded by the BNLF1 gene of the EBV. Each clone reacts with different epitopes on the hydrophilic C-terminus of the cytoplasmic domain of LMP-1. This antibody stains strongly with EBV-positive lymphoblastoid cell lines and EBV infected B cell immunoblasts in infectious mononucleosis. EBV, also designated human herpesvirus 4 (HHV-4), is a member of the herpesvirus family and is one of the most common human viruses. EBV infects B cells and, though often asymptomatic, it can cause infectious mononucleosis, a disease characterized by fatigue, fever, sore throat and muscle soreness. This antibody recognizes a protein of 145 kDa, identified as CD11c. CD11c (ITGAX), a member of the leukointegrin family, shares the same beta subunit with other members of the leukocyte adhesion molecule family, which includes CD11a (LFA-1), CD11b (MAC-1) and CD11d (ITGAD), but has a unique alpha chain. CD11c has been shown to play a role in phagocytosis, cell migration, and cytokine production by monocytes/macrophages as well as induction of T-cell proliferation by Langerhans cells. CD11c is expressed prominently on the plasma membranes of monocytes, tissue macrophages, NK cells, and most dendritic cells (DCs). A lower level of expression is also observed on neutrophils as a result of its high level of expression on most DCs. An antibody to CD11c may aid in identification of lesions with histiocytic origin. The protein encoded by this gene is a member of the keratin gene family.
