Protein phosphorylation is a fundamental event in the regulation of a large number of intracellular processes. Phosphorylation of specific tyrosine residues is the result of activation or stimulation of their respective protein tyrosine kinases. VISTA / GI24 is a transmembrane protein expressed in bone, on embryonic stem cells (ESCs), and on tumor cell surfaces. On ESC s, Gi24 appears to positively interact with BMP-4, potentiating BMP signaling and the transition from an undifferentiated to a differentiated state. On tumor cells, Gi24 both promotes MT1-MMP expression and activity and serves as a substrate for MT1-MMP. This increases the potential for cell motility. Mature human Gi24 contains a 162aa extracellular region with one V-type Ig-like domain and a 96aa cytoplasmic domain. Human Gi24 undergoes proteolytic cleavage by MT1-MMP, generating a soluble 30 kDa extracellular fragment plus a 25-30 kDa membrane-bound fragment. Recognizes a 53 kDa protein, identified as the Ornithine Decarboxylase (ODC-1). ODC is the initial and rate-limiting enzyme in the biosynthetic pathway of polyamines and is involved in the conversion of ornithine to putrescine. The biological activity of ODC-1 is rapidly induced in response to virtually all agents known to promote cell proliferation including hormones, drugs, growth factors, mitogens, and tumor promoters. Reportedly, ODC mRNA levels are elevated in lung carcinomas as well as in colon adenomas and carcinomas. ODC activity in colorectal carcinomas is greater than those in adenomas and normal mucosa. Zinc finger proteins contain DNA-binding domains and have a wide variety of functions, most of which encompass some form of transcriptional activation or repression. Zinc finger protein 690 (ZNF690), also known as ZSCAN29, is a 851 amino acid member of the Kruppel C2H2- type zinc finger protein family. This antibody recognizes a protein of 30-33 kDa, which is identified as CD20. It is a non-Ig differentiation antigen of B-cells and its expression is restricted to normal and neoplastic B-cells, being absent from all other leukocytes and tissues. CD20 is expressed by pre B-cells and persists during all stages of B-cell maturation but is lost upon terminal differentiation into plasma cells. This MAb can be used for immunophenotyping of leukemia and malignant cells, B lymphocyte detection in peripheral blood and B cell localization in tissues. It reacts with the majority of B-cells present in peripheral blood and lymphoid tissues and their derived lymphomas. It also has affinity for the active form of 92 kDa Type IV collagenase (also known as MMP-9 or gelatinase B). Spectrin is an actin crosslinking and molecular scaffold protein that links the plasma membrane to the actin cytoskeleton, and functions in the determination of cell shape, arrangement of transmembrane proteins, and organization of organelles. This antibody recognizes an epitope encoded by exon v4 on the variant portion of human CD44. The CD44 molecule belongs to a family of cellular adhesion molecules found on a wide range of normal and malignant cells in epithelial, mesothelial and hemopoiesis tissues. CD44 is a single gene with 20 exons, of which 10 are normally expressed to encode the basic CD44 (H-CAM) molecule. Cathepsin K is a lysosomal and secreted cysteine proteinase involved in bone remodeling and resorption. This protein, which is a member of the peptidase C1 protein family, is predominantly expressed in osteoclasts. However, the encoded protein is also expressed in a significant fraction of human breast cancers, where it could contribute to tumor invasiveness. Mutations in this gene are the cause of pycnodysostosis, an autosomal recessive disease characterized by osteosclerosis and short stature. PMEPA1 (prostate transmembrane protein, androgen induced 1 is a 287 amino acid single-pass membrane protein that contains WW-binding motifs and localizes to the cell membrane. Expressed at high levels in prostate, kidney and ovary, PMEPA1 interacts with NEDD4 and may play a role in regulating AR (androgen receptor) levels, specifically in prostate cells. Down regulation of PMEPA1 is observed in prostate tumors, suggesting that PMEPA1 may exhibit activity as a tumor suppressor. This antibody is a mixture of four different monoclonal antibodies. This antibody is specific to 60 kDa latent membrane protein (LMP-1) encoded by the BNLF1 gene of the EBV. Each clone reacts with different epitopes on the hydrophilic C-terminus of the cytoplasmic domain of LMP-1. This antibody stains strongly with EBV-positive lymphoblastoid cell lines and EBV infected B cell immunoblasts in infectious mononucleosis. EBV, also designated human herpesvirus 4 (HHV-4), is a member of the herpesvirus family and is one of the most common human viruses. EBV infects B cells and, though often asymptomatic, it can cause infectious mononucleosis, a disease characterized by fatigue, fever, sore throat and muscle soreness.
